人感染猪链球菌的病原学及分子生物学特征的研究

摘　　要：目的对人感染猪链球菌的病原学及其分子生物学特征进行研究。方法将疑似猪链球菌感染患者血液、脑脊液进行增菌培养,对所分离的病原菌进行染色形态观察、A P I Strep 20生化鉴定,并合成引物检测人感染猪链球菌特异性16SrRNA基因片段2、型猪链球菌特异性夹膜多糖编码基因片段cps2J、溶菌酶释放相关蛋白编码基因片段mrp、细胞外蛋白因子编码基因片段ef、溶血素基因片段sly以及甘油醛-3-磷酸脱氢酶基因片段gapdh。结果将患者脑脊液中分离的病原菌,经A P I20Strep生化鉴定编码为0640473,证实为猪链球菌2型,PCR扩增结果也证实是猪链球菌2型,基因测序结果与Genebank中注册的猪链球菌2型序列同源性在96%以上。结论人感染猪链球菌的病原学及其分子生物学特征符合猪链球菌2型。Objective Isolation streptococcus suis and identification the type of the strain. Methods The bloed and cerebro spinal fluid （CSF）were taken from a infected man, the samples were detected by blood nutrient agar medium,and causative organism were observed by the sharp of the organism, API Strep biochemical tests and PCR was used to detected genes. Results A strains of streptococcus were isolated from the CSF of the infectd man, with API 20 Strep test, the cede of the strain was 0640473, PCR also confomls that the strain of streptococcus was streptococcus suis Ⅱ . Conclusion Characteristics of molecular biology and aetiology being in accord.
