摘 要:目的:探讨异丙酚对大鼠肾脏缺氧/复氧损伤的影响。方法:将Wistar大鼠24只随机分为3组,对照组(A组)、缺氧/复氧组(B组)和异丙酚预处理组(C组)。采用抗原竞争ELISA法检测大鼠肾组织8-异前列腺素F2α(8-iso-PGF2α)水平,黄嘌呤氧化酶化学发光测定法检测其超氧化物歧化酶(SOD)活性。结果:缺氧复氧后,B组肾组织中8-iso-PGF2α水平明显升高,而SOD的活性则明显降低,与A组相比差异有非常显著意义(P<0.01);C组与B组相比,8-iso-PGF2α水平明显下降,而SOD活性明显上升,差异亦有非常显著意义(P<0.01)。结论:异丙酚能明显降低缺氧/复氧后肾组织的8-iso-PGF2α水平,提高SOD活性,从而增强机体清除超氧阴离子自由基的能力,抑制脂质过氧化,提示其对大鼠肾脏缺氧/复氧损伤具有明显的保护作用。Objective To investigate effects of propofol on renal hypoxia/reoxygenation (H/R) injury in rats. Methods Twenty-four Wistar rats were randomly assigned to the control group (group A), H/R group (group B) and H/R +propofol group (group C). The 8-iso-prostaglandin F2α(8-iso-PGF2α) levels in the renal tissue were determined by enzyme-linked immunosorbent assay (ELISA) and the superoxide dismutase (SOD) activity was detected by chemiluminescent analysis. Results As compared with those in group A, the 8-iso-PGF2α level in the renal tissue was significantly elevated in group B after H/R while the SOD activity was markedly lowered ( P 〈 0. 01 ). The level of 8-iso-PGF2α in group C decreased significantly and the SOD activity increased noticeably as compared with those in group B ( P 〈 0. 01 ) . Conclusion Propofol can enhance the body's ability to clear superoxide anions by lowering the level of 8-iso-PGF2α and elevating the SOD activity after H/R and can inhibit lipid peroxidation, which is suggestive of its role in protection against renal H/R injury in rats.
