摘  要:目的:研究低强度电磁场对皮肤表皮干细胞的分化与调控和对创面表皮再生的作用。方法:建立大鼠皮肤深II度烧伤的动物模型(共40只)。随机分为对照组(10只)及实验组(3组频率分别为1Hz,10Hz和50Hz脉冲磁场输出5mT,每组各10只)。收集创面组织标本,进行HE染色、β1整合素及角蛋白14(K14)免疫组化,观察皮肤表皮干细胞在创面愈合过程中的分布情况。并对比两组创面愈合率、愈合时间、创面感染率、毛囊数量。结果:治疗组创面愈合时间明显提前(P<0.01)。实验组间对比以50Hz组创面愈合时间缩短较明显(P<0.05),实验组创面愈合率与对照组对比有明显差异(P<0.01),实验组间对比,以50Hz组创面愈合率数值较大(P<0.01)。β1整合素和K14表达于基底细胞和毛囊细胞,实验组表达水平明显高于对照组(P<0.01),其中50Hz频段效果最为显著。结论:低强度电磁场能促进烫伤创面愈合,其机制可能是促进创面表皮干细胞增殖与分化。Objectives: To observe the effects of low intensive pulse magnetic field on the proliferation and differentiation of skin ESCs during wound healing process. Methods: After setting up rats' model of deep second-degree burn, 40 model rats were divided into control groups (10) and experimental groups (30) randomly. The pulse with frequency of 1 Hz, 10 Hz and 50 Hz were exerted on 10 rats separately in experimental groups. The output of pulsed magnetic fields (PEMFs) was 5mT. The wound specimen were collected and dyed with HE, and immunohistochemistry of integrin β1 and keratin 14 (K14), while the distribution of ESCs in wound was observed. The healing rate, healing time, infection rate of the wound and the quantity of hair follicles were compared among four groups. Results: The average wound healing time of therapy groups were significantly shorter than that of control group (P〈0.01), the expressions ofintegrin βt and keratin 14 (K14) of PEMF groups were obviously higher than that of control groups (P〈0.01), which was based on the basal cells and hair follicle cells. The frequency of 50Hz caused the most remarkable change than others. Conclusions: The reasons of low intensive pulse magnetic field to heal wound perhaps relate with the promotion of proliferation and differentiation of epidermal stem cells.
