摘  要:目的观察胚胎神经干细胞(neuralstemcells,NSCs)对成年大鼠脊髓损伤(spianlcordinjury,SCI)的修复及对轴突再生的作用。方法制作20只大鼠SCI(T7)模型,随机分为实验组和对照组。伤后2d实验组移植经全离培养的Wistar大鼠NSCs;对照组只注入DMEM培养液。移植后第1、2、4、6周经电镜及免疫组化观察移植对脊髓轴突再生的影响。结果实验组NSCs与宿主融合较好,损伤区可见幼稚的呈束状排列的再生轴突;对照组轴突变性未见再生轴突。结论NSCs植入异体大鼠打击伤后的脊髓可存活,并产生髓鞘样物质促进宿主脊髓轴突再生。Objective To observe the therapeutic effect of neural stem cells (NSCs) transplantation on spinal cord axonal regeneration. Methods Twenty rots were divided into two groups, 10 rats in each group. Two days after SCI models were made, NSCs suspension, purified from Wistar rats and transplanted into the injured spinal cords at T7 levels, while the vehicle solution (DMEM) was injected in the control gruop. At 1, 2, 4, 6weeks after transplantation, the samples were procured and studied by electron microscope and immunohistochemical method. Results The morphology of translated NSCs was almost normal, and NSCs arrangement was good in the host. Axons could regrow into injured spinal cords. The axons regeneration was not found in the controls. Conclusion Transplantation of NSCs could integrate with the host and could promote the axons regeneration of host.
