摘 要:目的探讨实验性豚鼠变应性鼻炎(Allergic rhinitis,AR)核转录因子-κB(Nuclear factor kappa B,NF-κB)和白介素-5(Interleukin-5,IL-5)的表达及相互关系,分析其在豚鼠AR发病中的作用。方法用免疫组化Elivision方法半定量测定脾脏组织NF-κB(亚单位P65)的活性细胞表达及双抗体夹心酶联免疫吸附技术(Enzyme labeledimmu-nosorbent assay,ELISA)定量测定IL-5的表达;比较豚鼠AR模型组及与正常组的差异。结果豚鼠AR模型组NF-κB P65阳性细胞比例和IL-5、嗜酸粒细胞(Eosinophil,Eos)明显高于正常组(P<0.05),且NF-κB P65阳性细胞比例与IL-5的表达呈正相关(r=0.85,P<0.05)。结论核转录因子-κB通过促进豚鼠AR发病过程中Th2类细胞因子IL-5的表达,进而增多鼻黏膜Eos的浸润而加重豚鼠AR症状。Objective To investigate the expression of NF-κB and IL-5 in spleen tissue of guinea pigs with experimental allergic rhinitis(AR) and the association between them to explore their possible implications in the pathogenesis of AR. Methods Immunohistochemical Elivision assay was taken to determine the expresfion of NF-κB subunit P65 and ELISA was used to detect the expression of IL-5 in spleen Tissue of guinea pigs with experimental AR. Then, a comparative study was carried out to see their expressing differences between modeling group and healthy one. Results The cell numbers with positive staining of NF-κB subunit P65, IL-5 and eosinophils (Eos) in nasal mucosa were significantly higher in the group of animals with experimental AR than those of healthy animals respectively (P〈 0.05), with a positive correlation in the cell number expressing NF-κB subunit P65 with that of IL-5 ( r = 0.85, P 〈 0.05). Conclusions NF-κB holds important pathogenetic implications in the pathological process of AR, via increasing the expression of Th2 cytokine IL-5 to promote Eos infiltrating in nasal mucosa to make AR manifestation exacerbated.
