摘 要:目的 探讨β-catenin在急性心梗后梗死区心肌组织的修复愈合过程中的表达变化。方法 建立大鼠急性心梗模型,分别用免疫组化和原位杂交方法检测大鼠急性心梗后心肌组织β-catenin的表达变化。结果 心肌梗死后1d,梗死区及非梗死区胞浆中均未见β-catenin蛋白的阳性表达;心肌梗死后4d,在梗死区的颗粒组织和边缘区部分细胞的胞浆中出现了β-catenin的阳性表达;7d时,其表达达高峰,颗粒组织的新生小血管内皮及平滑肌细胞也出现了明显的阳性染色;14d后β-catenin的表达渐减弱,直至消失。结论 胞浆中游离状态的β-catenin可能参与了诸如肌纤维母细胞样细胞和血管内皮细胞的增殖和迁移等过程;在急性心梗后心肌组织的修复过程中对颗粒组织以及新生血管的形成发挥了一定的作用。Objective To investigate the expression of β-catenin, a member of wnt-frizzled signal transduction pathway during wound healing process after acute myocardial infarction. Methods The left descending coronary artery of rats was tigated to induce myocardial infarction (MI) , immunohistoehemistry SABC method and in situ hybridization were performed to detect the expression of β-catenin. Results One day after MI, the β-catenin expression could not be detected; on day 4 after MI, β-catenin positive cells appeared in granulation tissue and the border zone of the infarct area; on day 7 after MI, the expression of β-catenin increased significantly and cytoplasmic β-catenin was observed even in the endothelial cells and smooth muscle cells of the newly formed and pre-existing blood vessels in the infarct area; in the tissue obtained 14 days after MI, the expression of β-catenin decreased gradually and could not be detected completely on day 28 after MI. Conclusion Cytoplasmic β-catenin may involve in the controlled proliferation and migration of myofibroblasts and vascular endothelial cells, which play a role during the wound healing process of formation of granulation tissue and neovaseularization after MI.
