摘  要:以欧美杨107(Populus×euram ericanacv.“74/76”)叶片为试材,采用根癌农杆菌介导法进行了胆碱单加氧酶(CMO)基因转化,获得了耐盐转基因植株。试验结果表明,15 mg.L-1卡那霉素(Km)可作为筛选转化细胞的选择压;在农杆菌工程菌液中添加200μmol.L-1的乙酰丁香酮(As)可以显著提高抗性芽诱导率;PCR及Southern检测证明CMO基因已整合进抗性植株基因组;组织培养条件下的植株耐盐性测定及离体叶片电导率测定结果证明,转基因植株的耐盐性得到了提高。Leaf disk of poplar( populus x euramericana cv. "74/76" ) used as material, agrobacterium tumefaciens-mediated transformation of Choline monooxygenase(CMO) gene was studied. The results confirmed that 15 mg·L^-1 Km could be used to select transgenic ceils. Adding 200μmolL^-1 AS into agrobactefium culture medium could increase Kan - resistant shoots rate. PCR assay and Southern blot analysis showed that CMO gene was integrated successfully into the genome of transformed plant. And salt tolerance test showed that transgenie plants grew better than non-transgenic plant on medium containing NaCl. The relative conductance of transgenic plants was less than that of non- transgenic under 0.5% NaCl stress.
