摘 要:目的:探讨表皮生长因子受体(EGFR)在紫外线诱导血管内皮生长因子(VEGF)分泌中的作用。方法:流式细胞仪检测EGFR和磷酸化EGFR的表达。ELISA检测上清中VEGF的水平。结果:紫外线照射后15min可以检测到磷酸化EGFR的表达,30min时达最高峰,2~8h降至基础水平。在整个过程中EGFR的表达量不变。(EGFR+\+)MEF细胞组VEGF分泌明显高于(EGFR+/-)MEF和(EGFR-/-)MEF细胞组,(EGFR-/-)MEF细胞组中波紫外线对VEGF分泌促进作用不明显。PD153035,可明显抑制VEGF分泌,1!m有抑制作用,5!m抑制作用增强。结论:EGFR是UVB诱导VEGF分泌信号途径中的关键靶点。Objectives: To explore the role of epidermal growth factor receptor (EGFR) on UV-induced VEGF secretion of MEF. Methods: Flow cytometry was used to detect total receptor and tyrosine-phosphorylated EGFR, respectively, the concentration of VEGF was determined by ELISA. Results: EGFR phosphorylation was detectable within 15 min after UV exposure. The level of EGFR phosphorylation reached peak 30 min after UV exposure, and was decreased to basal levels 2 to 8 h after UV exposure. VEGF expression was not increased in EGFR (-/-) MEF following UV exposure. In contrast VEGF expression, was significantly increased in egfr (+/+) MEF after UV exposure. PDI53035 inhibited the induction of VEGF protein expression in UVB-treated cells in a dosedependent manner. Conclusion: EGFR plays an important role in UV-induced VEGF secretion in MEF.
