摘 要:目的了解珠海地区蚊媒携带流行性乙型脑炎(乙脑)病毒情况,为控制乙脑在人群中的流行提供依据。方法采集成蚊进行鉴定,并对蚊悬液采用细胞培养、逆转录聚合酶链反应(RT-PCR)以及荧光定量PCR技术进行病毒鉴定,并进行克隆与序列测定。结果2005年全年在养猪场共捕获成蚊876只,经鉴定隶属4属8种,其中以三带喙库蚊(35.73%)、致倦库蚊(32.54%)和海滨库蚊(21.35%)为主。在珠海各口岸捕获的542只蚊样本中,则以白纹伊蚊为主,占82.47%,其次为致倦库蚊,占16.25%。应用荧光定量PCR技术,在珠海地区注册养猪场和口岸的蚊媒中检出1份乙脑病毒阳性样本,但强度较弱,这与珠海地区是乙脑低发地区相一致。结论珠海地区存在乙脑的主要传播媒介,并且在蚊媒中携带有乙脑病毒,提示不能放松对乙脑的防控工作。Objective To investigate species composition, distribution and the situation of mosquito vectors of Japanese encephalitis virus(JEV) in Zhuhai area, hereby to provide scientifical evidences for strengthening monitoring, preventing and controlling the Japanese encephalitis. Methods Mosquitoes were trapped at 4 hoggeries and ports in Zhuhai from january, 2005 to June, 2006, and provided by Zhuhai CDC. The samples of mosquito vectors were ground to make suspension, the RNA of virus were extracted directly, or after suspension was inoculated into cells of C6/36s and educated, and then RT-PCR was carried out; The positive products were cloned and sequenced. Results 876 adult mosquitoes were captured during 2005 in the registered hoggeries. They belonged to 4 genera and 8 species, of which Culicinae species were the most dominant species. Cx. tritaeniorhynchus accounted for 35.73 %, Cx. quinquefasciatus 32.54 % and Cx. sitiens 21.35 %. 542 adult mosquitoes were captured in the ports of Zhuhai. Among them, 82.47 % were Aedes albopictus, 16.25 % were Cx. tritaeniorhynchus. 1 positive sample was identified as a mild JEV by fluorometric PCR from the mosquitoes( Cx. sitiens) in the registered hoggeries and the ports of local area. It is accordant to the fact of low local epidemic JE in Zhuhai. Conclusion Despite of the low local epidemic of JE in Zhuhai, the main vectors of JE exist and are dominant in Zhuhai, the prevention and control of JE can't be overlooked.
