摘 要:目的:β肾上腺素能受体(β-adrenoceptor,β-AR)包括β1,β2和β33种亚型,分析其基因多态性与大动脉僵硬度的关系。方法:试验于2003-02/2004-09在重庆医科大学生物医学工程研究室进行。选择在重庆医科大学体检的健康受试者94名,男49名,女45名,对试验均知情同意。测试受试者平卧位的血压,然后计算脉压差(脉压差可反应大动脉僵硬度)。用聚合酶链反应-限制性片段长度多态性技术和等位基因特异性聚合酶链反应法分析β1-AR的Ser49Gly和Arg389Gly,β2-AR的Arg16Gly和Gln27Glu,β3-AR的Trp64Arg基因多态性,并分析其基因多态性与脉压差的相关性。结果:94名受试者全部进入结果分析。①β2-AR的Arg16Gly基因多态性与脉压差有显著相关性(P<0.05),其中Arg/Gly杂合子基因型携带者的脉压差值最大(49.64±9.99)mmHg(1mmHg=0.133kPa),Arg/Arg纯合子基因型携带者的脉压差值最小(43.70±9.88)mmHg,而Gly/Gly纯合子基因型携带者的脉压差值居中(46.00±12.12)mmHg。②β1-AR的Ser49Gly和Arg389Gly、β2-AR的Gln27Glu和β3-AR的Trp64Arg基因多态性与脉压差无关(P>0.05)。结论:β2-AR的Arg16Gly基因多态性与脉压差值有显著相关性,故而β2-AR的Arg16Gly基因多态性与大动脉僵硬度有相关性。AIM: To analyze the correlation between β-adrenoceptor (β-AR), including three subgroups: β1, β2 and β3, gene polymorphism and largeartery stiffness. METHODS: The experiment was conducted in the Biomedical Engineering Laboratory of Chongqing University of Medical Science from February 2003 and September 2004. Ninety-four healthy examinees (male 49; female 45) for physical examination were selected. All the people were informed and agreed to join the experiment. The blood pressures of the subjects in the supine were detected, and then the pulse pressure (reflected the stiffness of large artery) was computed. Polymerase chain reactionrestriction fragment length polymorphism (PCR-RFLP) and allele specificPCR techniques were used to determine the polymorphism of Ser49Gly and Arg389Gly of the β1-AR gene, Arg16Gly and Gln27Glu of β2-AR gene and Trp64Arg of β3-AR gene and explore the correlation with pulse pressure. RESULTS: All the 94 subjects were involved in the result analysis.① The Argl6Gly polymorphism of β2-AR gene was significantly associated with pulse pressure (P 〈 0.05). Individuals with Arg/Gly heterozygote had the biggest pulse pressure (49.64±9.99) mm Hg, with Arg/Arg homozygote had the smallest pulse pressure (43.70±9.88) mm Hg, and the individuals with Gly/Gly homozygote had the intermediate pulse pressure (46.00±12.12) mm Hg. ②No association was found between Ser49Gly, Arg389Gly, Gln27Glu and Trp64Arg polymorphisms with pulse pressure, respectively (P〉 0.05). CONCLUSION: Arg16Gly polymorphism of β2-AR gene is significantly associated with pulse pressure. Therefore, Arg16Gly polymorphism of β2- AR gene has correlation with large-artery stiffness.
