摘 要:目的:观察大鼠在不同运动强度一次性跑台运动后骨骼肌结构的变化情况,以及在运动后不同时相点血清总抗氧化能力、超氧化物歧化酶活性及丙二醛含量等指标的变化规律,并分析其相互关系。方法:实验于2004-10/2005-03在成都体育学院动物实验室及成都中医药大学附属医院检验科完成。选择成年健康雌性SD大鼠72只,随机数字表法分为空白对照组(n=8),中等运动负荷量组(n=32),高运动负荷量组(n=32);中等运动负荷量组、高运动负荷量组又分为运动后即刻,24,48,72h4个时相点,各时相点8只。中等运动负荷量组、高运动负荷量组都进行一次性跑台运动,其中中等运动负荷量组速度15~16m/min,坡度为0°,运动60min;高运动负荷量组速度19~21m/min,坡度为-16°,运动90min。运动后取股动脉血,检测血清总抗氧化能力、超氧化物歧化酶活性及丙二醛含量,并进行比目鱼肌电镜和光镜观察。结果:纳入动物72只,均进入结果分析。①中等运动负荷量组、高运动负荷量组大鼠运动后骨骼肌纤维结构发生明显改变,其变化以运动后24h较为严重,中等运动负荷量组出现的变化较高运动负荷量组轻微。②中等运动负荷量组运动后即刻,24,48h及高运动负荷量组运动后即刻血清总抗氧化能力水平较运动前显著增高[分别为(12.0±2.6),(13.1±2.6),(17.6±2.5),(12.0±2.1),(9.1±1.2)U/mL,P<0.01],中等运动负荷量组峰值出现在48h,高运动负荷量组出现在运动后即刻,高运动负荷量组在运动后48h和72h都显著低于中等运动负荷量组。③运动后血清超氧化物歧化酶活性都出现先降后升,中等运动负荷量组运动后即刻,24,48h及高运动负荷量组运动后即刻较运动前显著降低[分别为(1457.0±186.7),(1543.6±298.4),(1423.6±321.7),(1333.6±255.1),(1967.0±220.0)μkat/L,P<0.01],中等运动负荷量组最低值出现在48h,高运动负荷量组最低值出现在运动后即刻。④AIM: To observe the histological changes of skeletal muscle's structure in rats after once treadmill exercise at different intensity and the changing rules of serum total antioxidant capacity (T-AOC), superoxide dismutase (SOD) and malondialdehyde (MDA) in corresponding phase points, and analyze the relationship among these indexes. METHODS: The experiment was conducted in the Animal Laboratory of Chengdu Sports University and the Clinical Medical Laboratory of Affiliated Hospital of Chengdu University of Traditional Chinese Medicine between October 2004 and March 2005. Seventy-two SD female adult rats were selected and randomly divided into normal control group (group A, n=8), middle-intensity exercise group (group B, n=32) and high-intensity exercise group (group C, n=32). Group B and group C were divided into instant group, 24-hour group, 48-hour group and 72-hour group with 8 rats in each phase point Rats in group B and C were forced to have a treadmill exercise. Rats in group B were forced to run at the speed of 15-16 m/minute at the slope of 0° for 60 continuous minutes, and rats in group C were forced to run at the speed of 19-21 m/minute and at -16°for 90 continuous minutes. Blood samples were drawn from femora after exercise, and the serum T-AOC, SOD and MDA were determined. Histological changes of fiber in solens were observed finder optical microscope and transmission electron microscope. RESULTS: A total of 72 rats were involved in the analysis of results. ① The fibrous structures of skeletal muscle in rats of group B and C were changed obviously, and the changes were the greatest at 24 hours after exercise than other phases. The changes in group B were milder than those in group C. ②The level of serum T-AOC at the instant, 24 hours and 48 hours after exercise in group B and at the instant after exercise in group C were increased significantly than those before exercise [(12.0±2.6), ( 13.1 ±2.6), ( 17.6±2.5 ), ( 12.0±2.1 ), (9.1 ±1.2) U/mL
