补充雌激素对去势雌兔关节液与血清中透明质酸含量的影响

摘　　要：目的:观察补充雌激素对去势雌兔血清与关节液中透明质酸水平的影响。方法:实验于2004-07/12在内蒙古医学院动物实验室和核医学科进行。40只雌兔随机分3组:①正常对照组8只,不作处理,仅用于取材。②模型组16只,施行卵巢切除术(去势术)。③雌二醇组16只,去势术后第3天肌肉注射苯甲酸雌二醇0.5mg/kg,以后每周二、五给药2次。各组均于去势术后2,4,6,8周时抽取关节液和外周血并制成血清,用放射免疫检测法测定透明质酸水平。结果:40只雌兔进入结果分析。①关节液透明质酸水平:术后第8周时模型组低于正常对照组[(305.53±91.11),(430.78±86.94)mg/L,P<0.05];雌二醇组各时间点与正常对照组比较差异不显著(P>0.05)。②血清透明质酸水平:术后第8周时模型组高于正常对照组[(6.04±1.60),(4.23±0.67)mg/L,P<0.05];雌二醇组各时间点与正常对照组比较差异不显著(P>0.05)。③关节液透明质酸与血清透明质酸比值:术后第6,8周时正常对照组高于模型组和雌二醇组(105.64±14.58,72.17±18.79,91.64±14.96,P<0.01,0.05;102.25±17.75,51.71±16.36,69.45±13.25,P<0.01),雌二醇组高于模型组(P<0.05)。结论:①兔体内雌激素水平下降可影响关节内透明质酸的水平,可能是诱发骨关节病的因素之一。②适当、及时补充雌激素可以预防和治疗骨关节炎。③关节液和血液中透明质酸比值的变化与雌激素水平下降相关,可作为观察雌激素替代疗法是否取得预期治疗效果的一种指标。AIM： To observe the influence of estrogen tonifying on hyaluronate in serum and synovial fluid in ovariectomized female rabbits. METHODS： From July to December 2004, the experiment was performed at the Animal Laboratory and Department of Atom Medicine, Inner Mongolia Medical College. Forty female rabbits were randomly divided into 3 groups： ①normal control group （n=8）, only being used to get experiment materials; ②model group （n=16）, the rabbits were ovariectomized; ③ estradiol, group （n=16）, estradiol benzoate 0.5 mg/kg were given to rabbits at day 3 after ovarieetomy by muscle injection, and then given every Tuesday and Friday. Peripheral blood and synovial fluid were extracted at weeks 2, 4, 6 and 8 after ovariectomy, and made into serum. Level of hyaluronate was measured with radioimmunodetection. RESULTS： A total of 40 female rabbits were involved in the result analysis. ①Level of hyaluronate in synovial fluid： The level was lower in the model group than the normal control group at week 8 after operation [（305.53±91.11）, （430.78±86.94） mg/L,P 〈 0.05]. There was no significant difference between the estradiol group and normal control group at each time point （P 〉 0.05）. ②Level of hyaluronate in serum： It was higher in the model group than the normal control group at week 8 after operation [（6.04±1.60）, （4.23±0.67） mg/L,P 〈 0.05]. There was no significant difference between the estradiol group and the normal control group at each time point （P 〉 0.05）. ③Ratio of hyaluronate in synovial fluid to that in serum： It was higher in the normal control group than the model group and estradiol group at weeks 6 and 8 after operation （105.64 ±14.58,72.17±18.79,91.64±14.96,P 〈 0.01,0.05 ; 102.25±17.75,51.71 ±16.36,69.45±13.25,P 〈 0.01 ）.
