高氧液对扭转/复位睾丸生精细胞凋亡及Caspase-3活性的作用

摘　　要：目的探讨高氧液对扭转/复位睾丸生精细胞凋亡的影响及作用机制。方法30只青春期Wistar大鼠随机等分为3组,对照组(C组)行左侧睾丸扭转720°即刻复位;睾丸扭转/复位模型组(M组)和高氧液治疗组(T组)行左侧睾丸扭转720°、2h后复位;T组于睾丸复位前15min静脉输注高氧液,输注60min。术后48h获取左侧睾丸,流式细胞术分析生殖细胞凋亡状况和Caspase-3活性,常规染色观察病理学改变。结果与C组比较,M组生精细胞Caspase-3活性和凋亡峰值明显增高(P<0.01),各级生精细胞数明显降低(P<0.01),睾丸病理损害明显。与M组比较,T组睾丸以上改变明显减轻(P<0.01)。结论高氧液可能通过抑制Caspase-3介导的生精细胞凋亡而发挥抗扭转/复位睾丸损伤的作用。Objective To detect the influence and mechanism of hyperoxic liquid on apoptosis of spermatogenic cells induced by testicular torsion/detorsion. Methods Thirty adolescent male Wistar rats were randomized into three groups （n = 10 for each） ： control group （C group） ,model group （M group） and hyperoxic liquid treat group （T group）. In C group the left testis underwent a clockwise 720° torsion and then untwisted at once. In M and T groups the left testis underwent a clockwise 720° torsion for 2 h and followed by detorsion; hyperoxic liquid was dripped intravenously 15 min before the testicular detorsion and then kept 60 min in M group. Left testis was excised at 48 h after detorsion and assessed for cell apoptosis and caspase-3 activity by flow cytometry, and the morphological damages were observed respectively. Results Compared with those in C group,tile apoptotic peak of spermatogenic cell and caspase-3 activity of the testis in M group were increased, while the spermatogenic cells were decreased significantly （ P 〈 0.01 ） ; the pathological lesions of the testicle were obvious. The changes mentioned above were alleviated obviously in T group compared with those in M group （ all P 〈 0.01 ）. Conclusion Hyperoxic liquid has a preventive effect on the testicular injuries through suppressing caspase-3 activity and lowering the levels of spermatogenic cell apoptosis induced by testicular torsion/detorsion.
