摘 要:研究建立对连续超薄切片电镜图像的三维可视化方法。对正常及用SARS冠状病毒感染非洲绿猴肾细胞(Vero E6)样品进行连续超薄切片,通过透射电镜得到线粒体断层图像,用表面显示技术对线粒体进行三维重构显示。正常和SARS冠状病毒感染的Vero E6细胞中的线粒体在结构上有明显区别,揭示出SARS冠状病毒感染Vero E6细胞线粒体的病理改变。以此建立起电镜连续超薄切片线粒体图像三维可视化方法。A novel visualization method of 3D (three- dimension'd) mitochondria structures reconstructed from serial ultrathin sections transmission electron microscope (TEM) images which applies surface rendering technique was presented. In order to validate the method, mitochondria in control and the Veto E6 cells, which were infected with SARS Coronavirus, were prepared by serial uhrathin sections, imaged in electron microscopy then reconstructed from serial section images and the results were compared with each other. As a result, mitochondria in control and the Vero E6 cells infected with SARS have remarkable difference in 3D structures, that illuminates pathological changes of Veto E6 cells. The results also testify the visualization method is valid.
