降脂汤对高胆固醇血症大鼠肾系膜细胞表型的影响

摘　　要：目的探讨降脂汤对高胆固醇血症大鼠肾系膜细胞表型的影响和可能机制。方法高脂饲料饲喂大鼠,建立高脂模型,以来适可为对照,观察降脂汤对大鼠血脂、肾功能及肾脏病理学的影响。结果降脂汤可降低大鼠TG、TC、LDL和Scr,升高HDL;还能减轻肾小球系膜病变和细胞外基质产生,降低肾小球系膜区α-SMA和Ⅳ型胶原表达。结论降脂汤对大鼠脂质性肾损害的保护作用可能与其在降脂基础上抑制肾小球系膜细胞的增殖和表型转化有关。Objective：To evaluate the protective effect of Jiangzhitang on pbenotypic changes of glomerular mesangial cell in hypereholesterotemia rats and investigate its possible mechanism. Methods： Fat diet was given to rats to establish the animal model of hypercholesterolemia. The effect of Jiangzhitang on the changes of serum lipid, renal function, Mesangial cell index and Mesangial matrix index were observed in the hypercholesterolemia rats. And lescol was used as a control group. Moreover, irnmunnohistochemical stain for a-smooth music actin and type Ⅳ collagen were also performed in renal tissues. Results：jiangzhitang could reduced the serum level of TC,TG,LDL and Scr, while increase the level of HDL. Mesangial cell index and Mesangial matrix index were significantly reduced, accompanied with decreased exprossion of a-SMA and type Ⅳ collagen. Conclusion.Jiangzhitang has the protective effect, on kidneys of hypercholcstcrolemia rats. The mechanisms of its protection might be due to inhibiting glomerular mesangial cells from undergoing multiplication and phenotypic change.
