摘  要:目的:探讨ALA-PDT结合hTGF-β1基因转染对结肠癌细胞体外增殖的抑制作用。方法:以腺病毒为载体将hTGF-β1基因导入人结肠癌细胞株8W480后,行PDT处理,应用免疫组化法检测TGF-β1的表达情况。通过MTT法检测转染后细胞增殖的抑制情况,应用流式细胞技术检测细胞周期的变化。结果:转染hTGF-β1基因后,TGF-β1蛋白表达明显增强。与单纯行PDT相比,G0/S周期阻滞作用更为明显。对SW480的增殖抑制效果明显提高(p<0.05)。结论:转染hTGF-β1基因可明显增强ALA-PDT对体外培养的结肠癌细胞SW480的增殖抑制作用。Objective:To study the effect of ALA PDT combined with human TGF- β1 gene transfection on proliferation of colon carcinoma SW480 cells. Methods:SW480 cells were infected with TGF - β1 - contained recombinant adenovirus, and treated with ALA - PDT. The expression of TGF - β1 was detected by immunohistechemical method. The proliferation of SW480 cells was detected by MTT assay. The C0 - G1 and S fraction were assayed by flow cytometry. Resuits: There was increased expression of TGE - β1 in SW480 cell line with transgenosis treatment at protein levels. When treated with ALA - PDT combined with TGF -β1 gene transfection, the G0 - G1 phase fraction increased remarkably, and the ability of cell proliferation decreased significantly in SW480 cell line, compared with that of PDT group( p 〈 0. 05 ). Conclusion : The proliferation of the colon carcinoma SW480 cell line may inhibited significantly by ALA - PDT combined with transgenosis of hTGF - β1.
