摘 要:目的观察高压氧(HBO)对缺氧后中性多核粒细胞(PMN)粘附功能及血管内皮细胞分泌单核细胞趋化蛋白(MCP-1)的调控作用。方法用人内皮细胞株(ECV 304)与脑梗死患者PMN作模型,观察一定剂量HBO(0.15 MPa,1 h)对脑梗死患者不同发病时期PMN在ECV 304上粘附率的影响;同时,利用ECV 304作为模型,给予缺氧干预后,观察HBO对ECV 304分泌MCP-1的影响。结果(1) HBO使1周内脑梗死患者PMN与内皮细胞株ECV 304的粘附率由46.77%降低为30.54%(P<0.01),对脑梗死3周组及健康对照组无明显影响。(2)HBO处理组ECV 304上清液中MCP-1浓度与对照组及常氧高压处理组相比显著降低(P<0.01)。结论HBO可以通过抑制内皮细胞对MCP-1的分泌及降低PMN—ECV304的粘附来减轻脑缺血再灌注病灶的炎症损伤。Objective To observe the effect of HBO in adjusting the adhesive ability of PMN and the secretion of MCP-1 in endothelial cells after anoxia. Methods Sing ECV 304 and PMN in patients with cerebral infarction as models, changes of the adhesive ability of PMN on ECV 304 was observed during various periods of HBO treatment, and so was the change of the level of MCP-1 excreted by ECV 304. Results The adhesive ability of PMN was obviously reduced in the 7 d group after HBO treatment (P 〈0.01 ). The level of MCP-1 excreted by ECV 304 in the HBO group was significantly reduced, compared with that in the two control groups. Conclusions HBO may alleviate the injury caused by anoxia by reducing the adhesion of PMN-ECV 304 and depressing the excretion of MCP-1 in endothelial cells in patients with cerebral infarction.
