创伤早期伤口液对体外培养真皮多能干细胞增殖迁移特性的影响

摘　　要：目的探讨真皮多能干细胞在创伤修复启动中的作用。方法采用前期建立的技术分离、纯化并扩增真皮多能干细胞、真皮成纤维细胞和血管内皮细胞;利用四唑盐比色试验(MTT法)检测不同时间、不同浓度伤口液对上述细胞的影响。在此基础上,观察伤后1d伤口液对真皮多能干细胞贴壁力和迁移力等与创伤修复密切相关的指标变化。结果伤后不同时相不同浓度伤口液对真皮多能干细胞增殖均具有显著刺激作用,但以伤后早期的伤口液作用最为明显。成纤维细胞和血管内皮细胞对伤口液反应均显著弱于真皮多能干细胞。伤口液作用后真皮多能干细胞贴壁力和迁移能力较对照组细胞显著增加。结论真皮多能干细胞是创伤愈合,特别是创伤修复启动过程中的一种重要的间充质干细胞,对其深入研究有望为阐明创伤修复启动分子机制提供依据。Objective To explore the effect of dermal pluripotent stem cells on the initiation of wound repairing. Methods Dermal pluripotent stem ceils, fibroblasts and vascular endothelial cells were routinely isolated, purified and expanded. Wound fluids of 1^st, 2^nd ,3^rd and 4^th day were collected from 12 male Wistar rats to culture the three types of cells mentioned above and their responses to wound fluids were investigated by MTT assay. The abilities of attachment and migration of dermal pluripotent stem cells stimulated by wound fluid of 1^st day were observed. Results The wound fluids could promote the proliferation of dermal pluripotent stem cells, especially the early-phase wound fluids. The responses of fibroblasts and vascular endothelial cells to wound fluids were not so strong as that of dermal pluripotent stem cells. The abilities of attachment and migration of dermal pluripotent stem cells significantly increased after the stimulation of early-phase wound fluids, suggesting that the dermal multipotent stem cells responsed well to wound fluids. Conclusion Dermal pluripotent stem cells may play an important role in the process of wound healing. The significance of dermal pluripotent stem cells in the initiation of wound repair is worth further investigation.
