摘  要:目的研究白芷对脂多糖和D-氨基半乳糖诱导败血症性休克小鼠的保护作用。方法50只雄性ICR小鼠随机分为正常对照组,模型对照组,阳性对照组,给药组。给药组分别按300,100 mg/kg腹腔注射白芷提取物,阳性对照组按30mg/kg腹腔注射地塞米松;正常对照组和模型对照组腹腔注射等体积生理盐水,30 m in后,除正常对照组外,其余各组均腹腔注射脂多糖(36 mg/kg)和D-氨基半乳糖(800 mg/kg)的混合溶液,比较各组门冬氨酸氨基转移酶(AST),丙氨酸氨基转移酶(ALT)和肿瘤坏死因子(TNF-α)含量,同时每d观察小鼠的死亡率至第3天。结果100 mg/kg剂量给药组存活率达到50%,300 mg/kg剂量给药组存活率达到70%,两个剂量给药组降低脂多糖和D-氨基半乳糖诱导败血症性休克小鼠血清AST和ALT含量的升高(P<0.01),同时显著降低TNF-α含量(P<0.001)。结论白芷提取物是通过减少TNF-α的分泌,对肝损伤起保护作用,最终降低脂多糖和D-氨基半乳糖诱导败血症性休克小鼠死亡率。Objectlve To investigate the protective effect of Angelica dahurica Root on lipopolysaccharide (LPS) and D- gaiactosamine ( D - GAIN) induced septic shock in mice. Methods Male ICR mice were intrapefitoneally injected with the Angelica dahurica Root extracts or saline 30 min before intrapefitoneally injection of LPS (36 mg/kg)/D - GaIN ( 800 mg/kg). Lethality rate was observed once a day for 3 days. Liver damage was evaluated by measuring serum AST and ALT activities, and the serum level of TNF - α was also measured. Results Pretreatments of mice with the Angelica dahurica Root extracts at doses of 100 mg/ kg and 300 mg/kg increased survival rates to 50% and 70%, respectively. As an initial mechanistic study, the level of TNF - α in serum samples of mice pretreated with the Angelica dahurica Root extracts ( 100 mg/kg and 300 mg/kg) was significantly lower than that of control LPS/D - GaIN treated group. The Angelica dahurica Root extract significantly decreased the serum AST and ALT activities, compared with those of control LPS/D - GaIN treated group. Conclusion The Angelica dahurica Root extract shows the protective effect on LPS/D - GaIN inducing lethality through down - regulation of TNF - α and hepatoprotection.
