摘 要:目的:对氧自由基(OFR)在高压氧(NBO)引致氧惊厥发生过程中的作用及其机理作初步探讨。方法:105只SD雄性大鼠被随机分为常压空气组、高压常氧氮组、HBO组、VitC-HBO组、VitE-HBO组、SOD-HBO组、HBO惊厥组、VitC-HBO惊厥组、VitE-HBO惊厥组和SOD-HBO惊厥组共10个组。进舱前VitC-HBO组、VitE-HBO组、SOD-HBO组、VitC-HBO惊厥组、VitE-HBO惊厥组和SOD-HBO惊厥组大鼠分别于腹腔内注射VitC100mg/kg,VitE100mg/kg或SOD1万U/kg,其它动物被注入等量生理盐水。然后常压空气组动物置于舱内用空气持续通风20分钟后出舱;其余各组暴露压力均为600kPa(6ATA)。高压常氧氮织动物暴露在含氧4%的氮氧混合气中,HBO组、VitC-HBO组、VitE-HBO组、SOD-HBO组的动物则暴露在纯氧中分别停留20分钟后减压;而HBO惊厥组、VitC-HBO惊厥组、VitE-HBO惊厥组和SOD-HBO惊厥组动物则在高压下停留至有惊厥大发作后开始减压。减压采用匀速减压法,以10分钟减至常压。采用电子自旋共振(ESR)技术?Aim:To study the effect of oxygen free radicals(OFR)on the occurrence and process of hyperbaric oxygen(HBO)induced convulsions,and the possible mechanism involved.Methods:105 male SD rats were divided into 10 groups(10~11 in each group):the normal pressure air group,the normoxic hyperbaric nitrogen group、the HBO group,the Vit C-HBO group,the Vit E-HBO group,the SOD-HBO group,the HBO-convulsions group,the Vit C-HBO-convulsions group,the Vit E-HBO- convulsions group and the SOD-HBO convulsions group.The contents of OFR in the brain of rats in each group after respective exposure treatments were measured by means of electron spin resonance(ESR)technique.Pathological changes in the brain of rats and the latent period of oxygen convulsions for all the experimental groups were also observed.Results:The contents of OFR in the brain of rats in the HBO groups were significantly higher than those in the control group(P<0.01).The latent period of oxygen convulsions for the vitamin C-HBO convulsions group and vitamin E-HBO convulsions group were obviously prolonged(P<0.01).Electron microscopic examination revealed significant swelling of the nerve fibre and mitochondria with swelling and breaking of nuclear membrane,reduction of ribo- some and myelin disorder or severance in the HBO group.But there were no significant differences of the changes between the SOD-HBO and HBO groups(P>0.05).Conclusion:The OFR contents in the brain of rats could be increased by HBO exposure,and OFR was found to be involved in the process of oxygen convulsions.But the elevation of OFR content was not closely correlated with the pressure it-self.
