摘 要:目的:研制有机磷农药电化学传感器,建立测定有机磷农药的高灵敏度的分析方法。为了降低电化学传感器对有机磷农药检测的高过电位和提高检测的灵敏度,用纳米金溶胶与溶胶-凝胶壳聚糖/二氧化硅杂化复合膜构成复合固酶基质,将乙酰胆碱酯酶(AChE)固定于普鲁士蓝修饰的玻碳电极表面,制备了有机磷生物传感器。方法:在1 mmol·L^(-1)氯化乙酰巯基胆碱,0.1 mol·L^(-1)磷酸盐(pH 7.5)溶液中,研究有机磷电化学传感器与有机磷农药中的对氧磷反应前后响应电流的变化,建立电化学测定有机磷农药的分析方法。结果:该传感器在电位为+0.20 V(vs.SCE)条件下对对氧磷进行检测的线性响应范围为5.0×10^(-8)~5.0×10^(-5)g·L^(-1)检出限为2.0×10^(-8)g·L^(-1)。结论:建立了快速,灵敏的测定有机磷农药的电化学分析方法。Objective:To develop an electrochemical biosensor based on incorporation of aeetylcholinesterase and gold nanoparticals for the highly sensitive determination of organophosphorus pesticides. A novel amperometric biosensor for detection of organophosphorus pesticides has been fabricated with acetylcholinesterase (ACHE) immobilized by gold nanoparticals and chitosan/silica sol -gel organic -inorganic hybrid composite film on the surface of prussian blue modified glassy carbon electrode in order to reduce high over - potential and enhance sensitivity of the electrochemical biosensor for paraoxon. Methods: In 0. 1 mol·L^-1 phosphate buffer solution containing 1 mmol·L^-1 acetylthiocholine cloride, the current changes of the electrochemical biosensor fabricated was taken as the response, Results: The response of the biosensor fabricated was related linearly to the concentration of paraoxon in the range from 5.0×10^-8 to 5.0×10^-5g·L^-1 at +0,20V(vs. SCE) with a detection limit of 2, 0×10^-8g·L^-1 Conclusion:An electrochemical biosensor for the determination of paraoxon was developed with a fast response and high sensitivity.
