RP-HPLC法同时测定野菊花注射液中木犀草素-7-O-葡萄糖苷和蒙花苷的含量

摘　　要：目的:建立同时测定野菊花注射液中木犀草素-7-O-葡萄糖苷和蒙花苷含量的方法。方法:采用 RP-HPLC 法,C_(18)柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.04%磷酸水溶液,进行梯度洗脱:在0～20 min时 B 相从20%线性改变至30%,20～30 min时 B 相从30%线性改变至40%;流速:1.0 mL·min^(-1);柱温:室温;紫外检测波长:348 nm。结果:木犀草素-7-O-葡萄糖苷和蒙花苷浓度分别在0.2～4.2μg·mL^(-1)(r=0.9998)和2.1～41.2μg·mL^(-1)(r=0.9998)范围内与峰面积呈良好的线性关系(n=6),平均回收率(n=9)分别为99.3%和99.9%。结论:方法简便、快速、准确,重现性好,为野菊花注射剂提供可靠的质控方法。Objective： To establish a method for the determination of luteolin - 7 - O - glycoside and linarin in Yejuhua injection simultaneously. Methods： The reversed phase HPLC system consisting of a C,s column （250 mm ×4.6mm,5μm） and a mixture of acetonitrile and 0. 04% phosphate acid with gradient elution as the mobile phase was adopted. The gradient condition was 0 - 20 min, acetonitrile 20% - 30%, 20 - 30 min, acetonitrile 30% - 40%. The flow rate was 1.0 mL · min ^- 1. The absorbance was monitored at 348 nm. Results： The linear response range（n =6） was 0. 2 -4. 2μg·mL^-1（r =0. 9998） and 2. 1 -41.2μg·mL^-1 （r =0. 9998） ,respectively. The average recoveries（ n = 9） of luteolin -7 - O - glycoside and linarin were 99. 3% and 99. 9%, respectively. Con- elusion：The assay demonstrated that the method had adequate accuracy and selectivity to measure the concentrations of luteolin- 7 -O -glycoside and linarin in Yejuhua injection.
