摘  要:目的:建立测定兔血清中庆大霉素浓度的微生物法,为庆大霉素药代动力学研究提供测定方法。方法:以短小芽孢杆菌[CMCC(B)63 202]为检定菌,利用含抗生素血清在含菌培养基中产生的抑菌圈直径与其浓度的对数呈线性关系进行测定。结果:兔血清中庆大霉素浓度在0.05~5.00μg·mL^(-1)范围内呈良好线性关系(r=0.9993),最低检测浓度为0.02μg·mL^(-1),高、中、低浓度加标血清的相对回收率分别为99.6%,101.1%,98.8%,日内与日间的 RSD 均小于9.5%。结论:本方法操作简单、快速,具有较高的灵敏度、精密度和准确度,适应于庆大霉素药代动力学的研究。Objective:To develop a microbiological method for detecting gentamycin in rabbitg serum. Method:Using Bacillus pumilus [ CMCC (B) 63 202 ] as experimental bacteria, gentamycin concentration (C) was detected according to the linear relationship between logC and diameter of circle (6) that the bacteria was inhibited. Resuits:There was a good linear relationships between φ and logC( r =0. 9993 ) within the range of 0. 05 -5.00 μg·mL^-1. The lower limit of detection was 0. 02 μg·mL^-1. The relative recoveries were 99. 6%, 101.1% and 98.8% ,and RSD for intra - day and inter - day were both less than 9.5%. Conclusion: The method is simple, fast, sensitive and accurate, and suitable for the study on pharmacokinetic of gentamycin.
