HPLC测定柏子养心丸中五味子酯甲、五味子甲素和五味子乙素的含量

摘　　要：目的:建立用 RP-HPLC 法测定柏子养心丸中五味子酯甲、五味子甲素、五味子乙素的含量。方法:样品用甲醇超声提取,过滤,取续滤液,采用 RP-HPLC 法直接测定,用十八烷基键合相,甲醇-水-冰醋酸(71:29:0.1)为流动相,流速1.0mL·min^(-1),检测波长220 nm。结果:五味子酯甲、五味子甲素、五味子乙素分别在0.0046～0.460 μg(r=1.000);0.0054～0.540 μg(r=1.000);0.0055～0.553 μg(r=0.9998)范围内,质量与峰面积具有良好的线性关系。平均加样回收率(n=6)分别为94.2%～102.4%,99.1%～106.8%,98.3%～105.6%;RSD 分别为2.2%～3.8%,2.7%～4.1%,1.9%～4.0%。方法用于柏子养心丸中五味子酯甲、五味子甲素、五味子乙素含量的测定,RSD(n=6)分别为2.3%～4.0%,1.8%～3.5%,1.5%～4.2%。结论:方法具有灵敏度高、准确、快速和简便的特点,能用于柏子养心丸中五味子酯甲、五味子甲素、五味子乙素含量的测定。Objective ： To establish an HPLC method for the determination of schisantherin, deoxyschisandrin and γ - schisandrin in Baiziyangxin pills. Method：The sample was extracted with methanol through supersonic wave and then filtered. The determination was performed by HPLC on Shim - pack VP - ODS（ 150 mm ×4. 6 mm,4.6 μm） C18 column using methanol-water- acetic acid glacial（71： 29： 0. 1） as a mobile phase ,the flow rate was 1.0 mL · min^-1, the detection wavelength was 220 nm. Results：The linear range of schisantherin, deoxyschisandrin and γ - schisandrin were 0. 0046 -0. 460 μg（ r = 1. 000） ,0. 0054 - 0. 540 μg（ r = 1. 000） and 0. 0055 - 0. 553 μg（ r = 0. 9998）respectively. The average recovery of three components were 94. 2% - 102.4% ,99. 1% - 106.8% and 98. 3% -105.6%. Conclusion： The method is accurate, quick, sensitive and can be used for measurement of schisantherin,deoxyschizandrin and γ-schisandrin in Baiziyangxin pills.
