摘 要:目的 制备山羊脱钙骨基质(decalcified bone matrix,DBM),研究其物理和生物力学特性,为其作为理想的组织工程骨支架材料提供理论依据。方法 制备山羊完全脱钙骨基质,观察其色泽、质地、孔隙度等物理性状,并用扫描电镜观察其孔隙度,测量其孔径;通过测定吸水率评估其亲水性;用生物力学测定仪测定其不同压缩比率下的瞬时形变恢复率、最大形变恢复率及最大极限压缩强度。结果 标准化制备DBM呈乳白色,密度均匀,表面布有较多的微孔,扫描电镜观察其孔径范围为350~450(409.2±39.7)μm,吸水后膨胀,最大值吸水率达(419.0±44.4)%;生物力学测定显示,DBM位移是压缩强度的函数,DBM压缩20%和30%时其最大极限强度分别为(1.43±0.35)N和(1.62±0.51)N,瞬时形变恢复率分别为100%和(91.0±8.2)%。结论 完全脱钙骨基质孔径大,亲水性强,形变恢复好,利于细胞的黏附生长,是理想的组织工程支架材料。Objective To prepare corn acteristics and biomechanical properties in o sue engineering. Methods Completely dec pletely decalcified bone matrix (DBM) and study its physical charrder to afford theoretical base for DBM as ideal scaffold of bone tisalcified bone matrix from goat was prepared with the standard methods. Its physical characteristics including colour, texture, porosities were observed, and the hole diameter and porosity were detected under scanning electron microscope (SEM). The hydrophilicity of DBM was evaluated by calculating water absorption. Its transient recovery rate of deformation, complete recovery time of deformation and maximal compressive strength were determined with biomechanical approach. Results DBM is ivory white with unique density and redundant microporosities of which the average diameter is (409.2±39.7 )μm. The maximum of absorption time and absorption rate of DBM were 12 h and (419.0±44.4)%. The transient recovery rate of deformation was 100% and (91.0±8.2) % when DBM is compressed 20% and 30% respectively. Conclusion The properties of DBM are larger diameter and higher rate of microporosities, better hydrophilicity, easy recover of deformation and more suitable for adhesiveness and growth of breeding cells, which is the ideal scaffold material of bone tissue engineering.
