摘 要:目的探讨匹罗卡品致痫大鼠海马结构齿状回苔藓纤维发芽(MFS)各时间点的变化。方法采用氯化锂-匹罗卡品腹腔注射制成大鼠癫痫持续状态(SE)及颞叶癫痫模型,利用Timm组化染色法进行MFS观察研究。结果Timm染色可见到实验组大鼠SE24h时齿状回内分子层及CA3区下锥体层即有MFS现象;实验组不同时点与对照组大鼠CA3区和齿状回内分子层MSF评分间差异均有统计学意义(P<0·05)。结论匹罗卡品模型基本复制了人类颞叶癫痫的临床病理特征;诱导的SE可造成大鼠海马结构易损区的MFS增加,从SE后24h即开始出现MFS现象,且随时间的延长其程度加重。Objective To investigate mossy fiber sprouting (MFS) changes of hippocampus at different time points in experimental rats status epilepticus (SE) induced by Lithium - pilocarpine. Methods Lithium - pilocarpine was used to set up rat models with SE induced by Lithium - pilocarpine. In addition, Timm histochemical technique was adopted to study MFS. Results MFS in the stratum subpyramidale of CA3 subfield and inner molecular layer of dentate gyrus developed at 24h in trial group. A significance difference was found between trial group and control group in grade of MFS of the CA3 subfield and dentate gyrus at different time points ( P 〈 0. 05 ). Conclusion The temporal lobe epilepsy model of rats induced by pilocarpine can replicate the features of human temporal epilepsy ; SE in adult rats can cause increase of MFS in the vulnerable areas of hippocampus, the MFS can develop after 24h of SE and the grade of MFS increases with time/goes on.
