摘  要:[目的]探讨四环素对化学性急性肺损伤(ALI)的干预作用机制。[方法]将成年雄性SD大鼠随机分为对照组、急性呼吸窘迫综合征(ARDS)组、四环素预防组及四环素治疗组;经股静脉注射油酸(OA)复制大鼠ARDS模型,四环素预防组于注射OA前30min缓慢(5min)静脉注入盐酸四环素,治疗组在注射OA后5min静脉缓慢注入盐酸四环素,对照组仅注射等量的生理盐水。在注射OA后不同时间,测定肺湿、干重及其比值和肺系数,应用酶联免疫吸附实验测定不同时间血清中促炎细胞因子白细胞介素-1β(IL-1β)和抗炎细胞因子白细胞介素-10(IL-10)水平,并用放射免疫法测定炎前细胞因子血浆内皮素(ET)含量。[结果]注射OA后1、2、4h血浆中的ET及血清中的IL-1β和IL-10水平明显高于对照组(P<0.01);四环素预防和治疗组血浆ET及血清IL-1β水平均显著降低,IL-10含量提升。[结论]ET、IL-1β和IL-10在ALI的发生发展中可能具有重要作用;四环素对化学性ALI具有一定的干预作用,其机制之一可能与其能影响ALI发病过程的各种细胞因子的产生有关。[ Objective ] To investigate the molecular mechanism of the intervention effects of tetracycline on ARDS, a severe type of ALl, caused by chemicals. [ Methods ] Male adult SD rats were randomly divided into four groups: control group, ARDS group, tetracycline pre-administrated group and tetracycline treatment group; the tetracycline intervention groups were further subdivided into low dose( 25 mg/kg )and high dose( 50 mg/kg )subgroups. The animal model of ARDS was produced by injection of oleic acid via femoral vein. Tetracycline were injected at 30 minutes before oleic acid injection ( preadministrated groups )and 5 minutes after oleic acid ( treatment groups ) respectively; the control rats were injected with 0.9% sodium chloride solution only. The lung wet/dry ratio were measured pegiodically after oleic acid injection. IL-1, IL-10 and ET in serum were also measured by enzyme linked immunosorbent assay( ELISA ) and radioimmunoassay respectively. [ Results ] The levels of IL-1 β, IL-10 and ET were significantly increased after OA injection. Tetracycline could effectively reduce the levels of IL- 1β and ET in serum of ARDS rats, and enhance the content of IL- 10 in serum of ARDS rats. [ Conclusion ] The cytokines such as IL-1β , IL-10 and ET might play an important role in the pathogenesis of ARDS caused by chemicals such as OA. Tetracycline has a definite positive effect on ARDS, which might be related to its intervention effect on the production of cytokines.
