摘 要:目的研究“补阳还五汤”在体外培养中是否具有一定的神经元保护作用。方法本研究采用原代培养胎鼠脊髓神经元,分别将高浓度的补阳还五汤(20μl/ml)、低浓度的补阳还五汤(10μl/ml)与体外培养5d的原代脊髓神经元共同孵育24h,空白对照组不加中药只加入等量的培养液。选用100μmol/L的谷氨酸作用于细胞1h,建立谷氨酸诱导原代脊髓神经元调亡的细胞培养模型,采用酶学方法(MTT法)评价神经元损伤程度。结果补阳还五汤组神经元线粒体内的琥珀酸脱氢酶的活性明显地高于空白对照组,组间有极显著的差异(P<0·001)。结论“补阳还五汤”可以部分地抑制谷氨酸的兴奋性神经毒性作用。Objective To study the protective effect of Buyanghuanwutang on neurons cultured in vitro, Methods 100μmol/Lglutamate(Glu) was used to incubate with primary spinal cord neurons for 1 h,which were per-exposed to Buyanghuanwutang(20μl/ml,10μl/ml) up to 24h. MTT analysis for the activity of cellular mitochondria succinate dehydrogenase was used as an indicator of cell viability,which was determind by measuring the optical density at 490 nm with Elisa microplate reader. Results The activity of cellular mitcohondria succinate dehydrogenase of BUYANGHUANWUTANG group was much higher than that of the control group. Conclusions Buyanghuanwutang can partly rescue Glu-injured rat spinal cord neurons.
