摘  要:采用AFLP(Amplified fragment length polymorphism)技术对黄瓜(Cucumis sativus L.)品种‘津绿4号’的单倍体、二倍体和四倍体进行基因组DNA多态性比较。结果表明:(1)从35对引物扩增获得2188条60~500bp的条带,多态性位点仅有20个,占0.92%,其中22对引物组合扩增的不同倍性材料的AFLP指纹没有明显差异;(2)在多态性位点表现中,以二倍体的条带存在,单倍体和(或)四倍体的条带丢失为主,在四倍体中扩增出1条特异带;(3)与相应的二倍体相比,单倍体和四倍体有特异片段的消失和增加。Haploid,diploid and tetraploid polymorphisms of cucumber variety ‘Junlü 4' were compared by AFLP. It was shown that (1) 2 188 bands ranging with 604500 bp were amplified with 35 pairs of primers. Only 20 polymorphic loci were found which accounted for 0. 92% and AFLP fingerprints amplified from the materials with different ploidies with 22 of the primer pairs did not remarkably differ; (2) The expression of the polymorphic loci showed that the bands remained present in the diploid material and the bands become abscent in the haploid and tetraploid materials in most cases and one specific band were amplified from the teraploid material; (3) In contrast with the diploid material,the haploid and tetraploid materials got their specific bands lost or added.
