摘  要:目的通过放射免疫分析,研究铅对神经元腺苷酸环化酶活性的影响。方法对无血清原代培养3d的胚鼠皮层神经元中,进行不同程度的体外铅暴露,继之用放射免疫分析测定神经元细胞腺苷酸环化酶的活性。结果放免分析显示,铅抑制体外培养的神经元腺苷酸环化酶活性。结论铅对体外培养的神经元腺苷酸环化酶活性的抑制作用可能是铅损害记忆能力的机制之一,放免法测定切实有效。Objective To investigate the adverse effect of lead exposure on the adenylate cyelase activity in eml)ryonal rat neuron cultures with RIA. Methods l,ead chloride ( PbCl2 ) solutions were added to cultures (3days old) of cortical material from rat embryo to reach the final concentrations of 10^-8mol/L, 10^-6moL/L and 10^-4mol/L respectivcrly. Each group consisted of 8 specimens and a fourth group with zero exposure was used as controls. The specimens were cultured for 3 weeks further and the adenylate eyclasc activity as expressed by eAMP concentrations were determined with RIA. Results The cAMP concentrations in the euhures decreased as the lead ion conccntrati-ns incresed. Differences of cAMP concentrations among the groups werc all significant (P 〈 0. 01 ). Conclusion Inhibition of adenylate cyclase activity by lead was clearly demonstrated. It might be inferred that the inhibition played some role in the lead toxicity on memory.
