摘  要:目的:通过观察服用天麻钩藤饮后高血压肝阳上亢证大鼠下丘脑的蛋白质的改变,从蛋白质组学角度探讨天麻钩藤饮的降血压和改善肝阳上亢证症状的机制。方法:实验于2005-10/2006-06在中南大学湘雅医院完成。①动物与中药:自发性高血压大鼠20只,完全随机(随机排列表法)分为治疗组及对照组各10只;正常SD大鼠10只(正常组)。附子汤:由60g单味制附子组成,先将药物浸泡30min,煎煮两次后再将药液合并浓缩为含生药0.1g/mL。天麻钩藤饮:天麻15g,钩藤15g,石决明20g,黄芩10g,杜仲10g,牛膝10g。石决明先煎20min,天麻后下;煎煮两次后再将药液合并浓缩为含生药0.1g/mL。②高血压肝阳上亢证模型的复制:所有动物同时喂养,治疗组及对照组连续灌服附子汤共21d,复制成高血压肝阳上亢证模型(收缩压明显增高,饮水量增加,性情变得更易激怒为造模成功)。③对照组自第22天起改为连续灌服蒸馏水14d,治疗组自第22天起改为连续灌服天麻钩藤饮14d,均于第36天处死。正常组大鼠,连续灌服蒸馏水35d,于第36天处死。④用固相pH梯度双向凝胶电泳分离各组大鼠下丘脑的总蛋白质,经考马斯亮兰染色,PDQuest7.0软件分析。结果:30只大鼠均进入结果分析。①治疗后治疗组大鼠的血压明显下降,饮水量减少,性情变得温和。②大鼠的下丘脑蛋白质图像匹配率达94%~100%,且重复性较好,其蛋白质主要分布在等电点4~8,相对分子质量14400~75000。治疗后有67个蛋白质点表达上升2倍及以上,有19个蛋白质点表达下降2倍及以上。结论:天麻钩藤饮治疗后高血压肝阳上亢证大鼠血压降低、症状改善,这可能与其下丘脑蛋白质的改变有关。AIM: To explore the changes in hypothalamic protein of rats with hypertension of liver-yang rising treated by Tianma gouteng yin decoction, and probe into its mechanism of decreasing the blood pressure and ameliorating the symptoms of liver-yang rising from proteomics. METHODS: The experiment was conducted in the Xiangya Hospital of Central South University from October 2005 to June 2006. ① Experimental animals and Chinese medicine: Twenty spontaneous hypertensive rats were randomly divided into treatment group and the control group with 10 rats in each group, and another 10 healthy SD rats were taken as the normal group. Aconiti praeparatae decoction was composed of 60 g of single radix aconiti lateralis preparatatotal, which was soaked for 30 minutes and then decocted twice to made into condense fluid, containing 0.1 g/mL of crude drug. Tianma gouteng yin: 15 g of gastrodia rhizomen, 15 g of gambir plant, 20 g of abalone, 10 g of scutellaria, 10 g of eucommia bark and 10 g of achyranthis, radix. Gastrodia rhizome was added at 20-minute-decoction of abalone, and condensed decoction, which containing 0.1 g,/mL of crude drug was prepared after twice decoction. ② Duplication of models of hypertension of liver-yang rising: All animal were fed at the same time, and rats in the treatment group and the control group were drenched with Aconiti praeparatae decoction for 21 continuous days to be made into models of hypertension of liver-yang rising (Models were considered to be successful as the systolic pressure and water-taking greatly increased and being irritable).③ Rats in the control group were drenched with distilled water from the 22^nd day for 14 continuous days. Rats in the treatment group were drenched with Tianma gouteng yin from the 22^nd day for 14 continuous days, and all rats were executed at the 36^th day. Rats in the normal group were drenched with distilled water for 35 days and then executed at the 36^th day.④Hypothalamic total protein (TP) of rats in each group were separated
