摘  要:实验于2005-10/2006-05在成都中医药大学针灸推拿学院和四川大学华西基础医学与法医学院组织胚胎学和神经生物学教研室完成。选用健康雄性叙利亚金黄地鼠10只,按随机抽签法分为电针组和捆绑对照组,每组5只。动物经驯化后进入自由运转状态,在恒定黑暗第3天于近似昼夜时间9:00分别给予电针(固定动物后,选百会、长强穴,电压1~3V,频率10~20Hz,15min)和捆绑(固定动物,不予其他处理)刺激,2h后用免疫组织化学方法检测各组金黄地鼠视交叉上核内γ-氨基丁酸免疫阳性细胞并对此进行计数。10只动物全部进入结果分析,无脱失。与捆绑对照组比较,电针组动物视交叉上核单位面积内γ-氨基丁酸阳性神经元计数增加(109.450±11.445,66.520±21.119,t=3.9963,P<0.05)。提示于近似昼夜时间9:00进行针刺可使视交叉上核γ-氨基丁酸表达增加,说明在主观白天针刺可影响视交叉上核γ-氨基丁酸的表达。The experiment was performed in the School of Acupuncture and Message of Chengdu University of Traditional Chinese Medicine and the Department of Histology, Embryology and Neurobiology, West China School of Basic and Forensic Medicine of Sichuan University from October 2005 t6 May 2006. A total of 10 healthy male golden Syrian hamsters were divided randomly into electroacupuncture and sizing control groups (n=5). Golden hamsters moved freely following domestication. On the third day of constant darkness, hamsters were treated by electroacupuncture [after fixing, Baihui (Du20) and Changqiang (Du 1), 1-3 V voltage and 10-20 Hz frequency, 15 minutes] and sizing (fixed the animals, without other disposal) at 9:00. At hour 2 following treatment, gamma aminobutyric acidlike immunoreactive neurons in hamster suprachiasmatic nucleus were observed by using immunohistochemistry and counted. Totally 10 hamsters were involved in the result analysis, no dropout: The expression of gamma aminobutyric acid in suprachiasmatic nucleus increased obviously in electroacupuncture group as compared with sizing control group [ 109.450± 11.445,66.520±21.119,t=3.996 3 ,P 〈 0.05 ). It indicated that the expression of gamma aminobutyric acid in suprachiasmatic nucleus could be increased by electroacupuncture at 9:00 day and night. The result showed that electroacupuncture could affect the expression of gamma aminobutyric acid in suprachiasmatic nucleus in the subjective day.
