复方布洛芬软胶囊的人体药动学和生物等效性

摘　　要：目的:研究复方布洛芬软胶囊的人体药动学和生物等效性。方法:采用随机交叉试验设计,20名健康男性受试者分别单剂量口服布洛芬软胶囊(布洛芬400 mg,参比制剂)和对乙酰氨基酚片(对乙酰氨基酚500 mg,参比制剂)与复方布洛芬软胶囊(布洛芬400 mg和对乙酰氨基酚325 mg,受试制剂), LC-MS／MS法和HPLC-UV法测定血浆中布洛芬和对乙酰氨基酚的浓度。结果:参比制剂和受试制剂布洛芬的cmax分别为(42±s 9)和(41±10)mg·L-1;tmax分别为(0．6±0．2)和(0．5±0．3)h;t1／2分别为(2．0±0．3)和(2．1±0．3)h;AUC0～12分别为(126±24)和(131±25)mg·h·L-1。对乙酰氨基酚的cmax分别为(7．5±1．2)和(4．6±0．9)mg·L-1;tmax分别为(0．8±0．5)和(0．8±0．4)h;t1/2分别为(2．8±0．6)和(3.0±0．7)h;AUC0～12分别为(27±6)和(17±4)mg·h·L-1。受试制剂布洛芬的相对生物利用度为(105±12)％,经剂量校正后,受试制剂对乙酰氨基酚的相对生物利用度为(96±12)％。结论:受试制剂与参比制剂具有生物等效性。AIM： To study the pharmacokinetics and bioequivalence of compound ibuprofen soft capsules in healthy volunteers. METHODS： Under a randomized crossover trial design, 20 healthy male volunteers were given separately a single oral dose of ibuprofen 400 mg plus acetaminophen 500 mg （reference preparation） and 400 mg/325 mg ibuprofen/acetaminophen （test preparation）. Concentrations of ibuprofen and acetaminophen in plasma were determined by methods of LC-MS/MS and HPLC-UV. RESULTS： The main pharmacokinetic parameters for ibuprofen of reference and test preparations were as follows： cmax （42 ± s 9） vs （41 ± 10） mg·L^-1：tmax （0.6±0.2） VS （0.5±0.3） h; t1/2 （2.0±0.3） vs （2.1±0.3） h; AUC0-12 （126±24） VS （131 ±25） mg· h· L^-1, respectively. The main pharmacokinetic parameters for acetaminophen of reference and test preparations were as follows： Cmax （7.5 ± 1.2） vs （4.6 ± 0.9）mg·h·L^-1 tmax （0.8 ± 0.5） vs （0.8 ± 0.4） h; t1/2 （2.8 ± 0.6） vs （3.0 ± 0.7） h; AUC0-12 （27 ± 6） vs （17 ±4） mg·h·L^-1, respectively. The relative bioavailability of ibuprofen was （ 105 ± 12） % and that of acetaminophen was （96 ± 12） % after dosage emendation for test preparation. CONCLUSION： The test and reference preparations possess bioequivalent.
