摘 要:本文使用CM-52离子交换树脂层析技术研究了明胶组分分离的有关因素,结果表明:氢键裂断剂和明胶溶液入柱前的处理对a组分的收率具有明显的影响,而一些操作因素,如离子交换时的介质酸度、缓冲洗脱液的组成、离子强度以及洗脱速度对过程的分辨率都具有明显的影响;此外,还发现,每批次处理的明胶量应与层析柱的离子交换量相称,否则将会影响分离的效果。In this paper,some factors to separate gelatin components were studied with the CM-52 ionic ex-change chromatography. It is indicated that the collecting rate of α-component was affected obviously by the breakup of hydrogen-bond and the treatment of gelatin before pumping into column, and other factors also influenced the separating resolution, for example: the acidity of medium in ion exchange, the composition of buffer solution, ion intensity and eluting velocity. It was shown that the amount of gelatin was equivalent to that of ionic exchange chromatography in batches, otherwise it changed the separating results.
