摘  要:以3年生柳桉优树环割促萌枝条为外植体。以MS为基本培养基,通过激素种类与浓度、大量元索配比等试验,探讨了柳桉腋芽诱导、继代增殖、生根培养基配方。结果表明:外植体宜采用中等幼嫩的茎段,用75%酒精消毒30s、0.1%升汞溶液消毒4min效果最佳;在3/4MS+0.3mg·L^-1 6-BA+0.2mg·L^-1NAA培养基上进行侧芽诱导。柳桉外植体出芽率最高;在改良MS+0.2mg·L^-1 6-BA+0.2mg·L^-1 NAA继代培养基上,柳桉增殖效果最好,其芽体健壮,生长正常,增殖率3倍以上;应用1/2MS培养基附加ABT 1^# 0.5mg·L^-1和IBA0.1mg·L^-1,其生根率可达95%以上。生根3~4条。苗木生长健壮。移栽成活率可达92%以上。这些配方已成功地应用于柳桉优良无性系苗木的工厂化生产,单个超净台月苗木生产能力达7万株以上。A tissue culture protocol by shoot proliferation is developed for Eucalyptus saligna through adjusting the plant growth regulators and ratio of macro-elements using sprouts coUected from stubs of three-year-old plus trees as explants. The main results are as follows: 1. Half lignified shoots were most suitable for explants. Dipping them in solution of 75 % alcohol for 30 seconds and then in 0.1% HgCl2 for 4 minutes are the best way for their disinfection; 2.3/4 MS supplemented with 0.3 mg·L^-1 6-BA, 0.2 mg·L^-1 NAA and 20 g·L^-1 sugar is crucial for shoot germination and proliferation, while amended MS supplemented with 0.2 mg·L^-1 6-BA, 0.2 mg·L^-1 NAA and 30 g·L^-1 sugar for dump shoot induction; 3.1/2 MS supplemented with 0.5 mg·L^-1 ABT-1 and 0.1 mg·L^-1 IBA is suitable for rooting, with rooting rate up to 95 % ; and 4. The survival rate of out-planting in nursery is more than 92 %, The protocol has been successfully used in large-scale clonal production of the seedlings.
