摘  要:探索高速、快速的玉米SOD分离与纯化方法,旨在为玉米SOD的规模化生产提供依据。利用金属螯合亲和层析和DEAE-纤维素离子交换层析相结合的方法分离纯化玉米SOD,得到2种达到电泳纯的SOD组分(A、B),并对它们的基本性质进行了初步分析。SDS-PAGE法测定A、B两组分的亚基分子量分别为20 kD和16.4kD;由电泳后的定位染色情况可知,这2种SOD组分均属于Cu/Zn-SOD;等电聚焦表明,A、B两组分的等电点分别为7.2和6.9。Probing a high - efficient and simple method is to provide the basis for the massive production of maize SOD. Isolation and purification of two SOD components (A and 13) from maize by metal chelate affinity chromatography and DEAE - cellulose ion - exchange chromatography and determination of their qualities had been carried out in this experiment. The analysis showed that the subunits molecular weights of two SOD components (A and 13) were 20 k D and 16, 4 k D respectively by SDS- PAGE;the types of the two SOD components were both Cu/Zn - SOD by the location staining method;and the isoelectric points (pI) of the two SOD components (A and B) were 7.2 and 6.9 respectively by isoelectric focusing.
