摘 要:目的探讨阿片受体在重复电针灸预处理诱导大鼠脑缺血耐受形成中的作用。并研究可能参与作用的阿片肽种类。方法雄性SD大鼠42只,随机分为空白对照组,戊巴比妥钠组,电针(EA)组,EA+δ受体拮抗剂naltrindole(NTI)组,EA+κ受体拮抗剂nor-binaltorphimine(BNI)组,NTI组和BNI组。EA组、EA+NTI组和EA+NBI组接受相同电针灸刺激(疏密波2/15Hz,1mA,30min/d)连续5d,EA+NTI组和EA+BNI组分别于每次电针灸开始前给予NTI(1mg/kg)或BNI(2.5mg/kg)腹腔注射;NTI组和BNI组单纯腹腔注射相同剂量的NTI或BNI。最后一次处理24h后采用MCAO模型。再灌注24h进行神经学评分后取脑,经TTC染色计算脑梗死容积。结果脑梗死容积EA组明显小于空白对照组(P=0.000);EA+NTI组与空白组比较没有明显的统计学差异(P=0.219),而与EA组有明显的统计学差异(P=0.000);EA+BNI组与空白组比较有明显统计学差异(P=0.000),与EA组比较没有明显的统计学差异(P=0.319);说明NTI(1mg/kg)能够阻断EA预处理诱导的脑保护效应,而BNI(2.5mg/kg)不能。结论阿片受体参与了重复电针灸刺激大鼠百会穴诱导的脑缺血耐受效应,δ受体发挥了主要作用。Objective To explore the role of opioid receptors in formation of cerebral ischemic tolerance induced by repeated electroacupuncture(EA) in rats. Methods Forty-two male SD rats were randomly divided into 7 groups(n=6 each): control group, pentobarbitone group, EA group, EA+ naltrindole(NTI)group, EA+ norbinaltorphimine (BNI) group, NTI group and BNI group. EA, EA+NTI and EA+BNI groups received electroacuuncture at the Baihui acupoint for 30 min a day for 5 days; NTI (lmg/kg) and BNI (2.5mg/kg) were intraperitoneally administered before the electroacupuncture stimulation in EA+ NTI group and EA+BNI group respectively. NTI group and BNI group only received NTI (lmg/kg) and BNI (2.5 mg/kg) respectively. Twenty-four hours after the last treatment, the right middle cerebral artery was occluded for 120 min. After 24 h reperfusion, the animals were sacrificed and the infarct volume of the brain was determined with TTC staining. Results Cerebral infarct volume: EA group presented smaller cerebral infarct size as compared with the control group (P=0. 000) ; there was no significant difference between the infarct volume of EA+NTI and control groups (P= 0. 219); EA+BNI group showed significant difference in infarct size compared with control group (P= 0. 000), while had infarct volume similar to that of EA group(P=0. 319). The results showed that NTI (1mg/kg), a δ-opioid receptor antagonist, when given before EA preconditioning, abolished ischemic tolerance induced by EA, while BNI (2.5mg/kg), a κ-opioid receptor antagonist, could not. Conclusions The present study suggests that δ-opioid receptor plays a role in ischemic tolerance induced by repeated EA in rats.
