强心甙类药对长春新碱抗肿瘤活性的协同作用

摘　　要：目的探讨强心甙类药物洋地黄毒甙或地高辛对长春新碱抗肿瘤活性的协同作用。方法台盼蓝排染法测定药物对Ｋ５６２细胞的杀伤作用；用小鼠肝癌腹水瘤Ｈ２２模型，观察药物对小鼠生命延长率的影响。结果洋地黄毒甙、地高辛能显著杀伤Ｋ５６２细胞，并有剂量依赖性，ＩＣ５０分别为０．０５９，０．１０１μｇ／ｍｌ；低浓度洋地黄毒甙或地高辛对低浓度长春新碱杀伤Ｋ５６２细胞呈现增强作用（ｑ＞１）。洋地黄毒甙或地高辛对长春新碱抑制Ｈ２２腹水瘤也呈增强作用（ｇ＞１）。结论洋地黄毒甙、地高辛对长春新碱抗肿瘤活性有增强作用。To study the synergetic effects of digitoxin and digoxin that belong to cardidac glycosides on antitumor activities of vincristin.Methods The cytotoxic effects of digitoxin or digoxin and their combinations with vincristin against K562 cells were determined by means of the trypan blau dye exclusion test.The influences of digitoxin or digoxin and their combinations with vincristin in lifespan in mice inoculated with hepatic ascites carcinoma cells(H22)were determined.Results Digitoxin or digoxin was able to kill K562 cells significantly with dose dependence.IC50 was 0.059,0.101μg/ml respectively.At low doses,the synergitic effect of digitoxin or digoxin on the cytotoxic effect of vincristin against K562 cells was shown.It was also shown that digitoxin or digoxin had synergetic effect on the inhibiting action of vincristin against hepatic aseites carcinoma (H22).Conclusion That digitoxin or digoxin may have synergetic effect on the antitumor activity of vincristin.
