摘 要:目的:研究山茱萸环烯醚萜总苷(ICO)对高糖或糖化终产物(AGEs)培养肾小球系膜细胞(GMC)增殖和Na+,K+-ATP酶活性的影响。方法:用含AGEs或高糖的培养液配制ICO、氨基胍,同时设对照。GMC培养48h后MTT法考察GMC的增殖情况,试剂盒测试细胞悬液中细胞蛋白含量,再进行Na+,K+-ATP酶测定。结果:于葡萄糖或AGEs中GMC的吸收度值增大,Na+,K+-ATP酶活性显著降低,而加入ICO后,则表现为对抗高糖或AGEs对GMC的增殖作用,并能提高Na+,K+-ATP酶活性。结论:ICO能抑制高糖或AGEs对GMC的过度增殖,修复高糖或AGEs致GMC的Na+,K+-ATP酶活性降低。Objective: To observe the influence of iridosides of Comus ofi%inalis (ICO) on the proliferation and Na^+,K^+-ATPase activity of glomerular mesangial cells ( GMC ) cultured in high glucose or advanced glycation end products (AGEs) medium. Methods : GMC were incubated in culture medium containing high glucose or AGEs in the presence of ICO and aminoguanidin for 48 hours. At the same time, the control and model groups were added. The proliferation of GMC were analyzed by MTT method. The Na^+,K^+-ATPase activity were tested by Na^+,K^+-ATPase test Kit. Result :GMC proliferated remarkably in glucose or AGEs medium in comparison with that in 5% FBS RPMI-1640 culture medium, and the Na^+,K^+-ATPase activity remarkably decreased. Whereas the ICO were added, the excess proliferation of GMC can be mostly reversed, and theNa^+,K^+-ATPase activity shown a significant restoration in comparison with that in model groups. Conclusion: ICO can inhibit the excess proliferation and restore the decreased Na^+,K^+-ATPase activity of GMC induced by high glucose or AGEs.
