霉酚酸酯对大鼠肾毒血清肾炎肾小球肝细胞生长因子与转化生长因子-β_1表达的影响

摘　　要：目的探讨霉酚酸酯对大鼠肾毒血清肾炎肾小球肝细胞生长因子(HGF)与转化生长因子-β1(TGF-β1)表达的影响。方法大鼠分为正常对照组、模型组、霉酚酸酯治疗组,每组各10只。治疗组给予霉酚酸酯20mg/(kg·d)灌胃,2周后测定各组24h尿蛋白、血肌酐、尿素氮水平,应用免疫组化方法观察肾小球HGF与TGF-β1表达。结果与模型组相比,霉酚酸酯治疗组能明显减少24h尿蛋白、血肌酐、尿素氮水平(P<0·01),显著增加肾毒血清肾炎大鼠肾小球HGF表达(P<0·01),减少TGF-β1表达(P<0·01)。结论该研究提示促进肾小球内HGF表达、抑制TGF-β1生成可能是霉酚酸酯减轻肾毒血清肾炎肾组织损伤程度,抑制新月体形成的机制之一。Objective To investigate the effects of mycophenolate mofetil （MMF） on the expression of hepatocyte growth factor （HGF） and transforming growth factor β1（TGF-β1 ） in glomerulus of rats with nephrotoxic serum nephritis. Methods Rats were randomly divided into control group, model group and MMF treating group [ Model group treated with MMF 20 mg/（ kg · d） ,by gavage] , 10 rats in each group. After 2 weeks, 24 h urinary protein, serum creatinine （Scr） , boold urea nitrogen （BUN） were measured, expression of HGF and TGF-β1 was determined by immunohistochemistry method. Results The levels of 24 h urinary protein, Scr, BUN in MMF treating group were significantly reduced in contrast with model group（ P 〈 0. 01 ）. HGF expression remarkably increased（P 〈 0. 01 ） and TGF-β1 expression obviously decreased（ P 〈 0. 01 ） in glomerulus of nephrotoxic serum nephritis compared with model group. Conclusion MMF could ameliorate renal injury and suppress crescent production ,which mechanism perhaps is to promote HGF expression and depress TGF-β1 expression of glomerulus.
