摘 要:目的观察虎杖苷(PD)对大鼠胃缺血再灌注损伤(GIRI)的保护作用及与抗氧化的关系。方法用小动脉夹夹闭腹腔动脉30min,复灌60min的方法建立大鼠GIRI模型,观察各组胃黏膜损伤指数及胃组织脂质过氧化物(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、一氧化氮(NO)和一氧化氮合酶(NOS)含量的变化。结果PD可明显减轻损伤模型中的胃黏膜损伤指数;能明显降低胃组织MDA含量,增加胃组织SOD、GSH-Px、NO、NOS水平。结论PD通过抑制胃缺血再灌注时氧自由基产生,提高组织抗氧化能力,对GIRI具有保护作用。Objective To research the protective effect of polydatin (PD) on gastric ischemia reperfusion injury in rats and relationship with antioxidative effect. Methods The gastric mucosa injury was produced in rats by clamping the celiac artery with a small clamp for 30 min and reperfusion for 60 min, then the rats were killed. Injury index were calculated. Methylenedioxyamphetamine ( MDA ) , superoxide dismutase ( SOD ), glutathione peroxidase (GSH-Px) , nitric oxide (NO) , nitric oxide synthase (NOS) of the gastic mucosa were detected. Results PD could remardably reduce injury index and the levels of MDA, significantly increase NO levels and the activities of SOD, GSH-Px, NOS. Conclusion PD can prevent acute gastric mucosa injury caused by ischemia reperfusion in rats, which may be related to inhibiting production of oxygen free radical during gastric ischemia reperfusion and enhancing the anti-oxidizing ability of tissues.
