摘  要:目的探讨腹腔注射1,6-二磷酸果糖(FDP)对大鼠惊厥持续状态(status convulsion,SC)所致脑损伤是否具有保护作用及可能机制。方法健康成年大鼠45只,按实验所需随机分为3组:FDP组、预使用FDP组和SC对照组,每组15只。经腹腔注射氯化锂-匹罗卡品(lithium-pilocarpine)诱发30 min的SC发作,检测3组大鼠脑组织病理改变,脑组织匀浆超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及血清神经元特异性烯醇化酶(NSE)含量的变化。结果(1)FDP组、预使用FDP组CA3区神经元坏死率分别为30.5%、31.5%,较SC对照组48.3%显著降低(P<0.01)。(2)SC后,脑组织匀浆SOD活性降低,MDA含量及NSE含量升高,FDP组、预使用FDP组较SC对照组SOD活性升高,MDA及NSE含量降低(P均<0.01)。(3)FDP组、预使用FDP组上述指标无显著性差异(P>0.05)。结论腹腔注射FDP可通过减轻氧自由基引起的脑损伤对SC大鼠脑组织起到保护作用。Objective To observe the protective effect of Fructose-1, 6-diphosphate (FDP) on rat brain damage after status convulsion (SC) and to explore the underlying mechanism. Methods Forty-five adult rats were divided randomly into FDP group, FDP pre-conditioning group and convulsion control group. Status convulsion (over 30 minutes) was induced in adult rats by intraperitoneal injection with lithium-pilocarpine. The pathological changes in cerebral tissues were observed. The biological activity of superoxide dismdese (SOD) and content of maleic dialdehyde (MDA) in the cerebral tissue were detected together with serum neuron-specific enolase (NSE) level. Results ① Neuronal necrosis rate in hippocampus CA3 region of FDP group and FDP pre-conditioning group were 30.5 % and 31.5 %, which were lower than that in convulsion control group(48.3% and P〈0.01). ② After SC, the biological activity of SOD was decreased but the contents of MDA and NSE were increased. Compared with convulsion control group, SOD levels in FDP group and FDP pre-conditioning group were higher, and contents of MDA and NSE were lower (P 〈 0.01). ③There was no significant difference between FDP group and FDP pre-conditioning group (P 〉 0.05). Conclusion By means of eliminating oxygen free radicals, FDP exerts protective effect on brain damage of rat status convulsion model.
