摘  要:目的:将兔成骨细胞(ROB)与血管内皮细胞(RVEC)联合培养复合肌瓣预构血管化骨。方法:将传代后的两种细胞冻存、复苏。制作体积为2.5 cm×1 cm×1 cm外消旋聚乳酸(PDLLA)支架并用Ⅰ型胶原包埋,收集复苏传代细胞,9只实验兔分3组,A组左侧背阔肌植入ROB与RVEC比例为2∶1细胞支架复合体,右侧植入ROB/PDLLA;B组左侧为ROB+RVEC/PDLLA、右侧为PDLLA;C组左侧为ROB/PDLLA、右侧PDLLA。2周、4周、8周观察体内成骨与血管化情况。结果:在体内预构血管化骨的实验中,两种细胞培养复合支架组在体内成骨能力及血管化程度均高于单纯成骨细胞复合支架组,而单纯支架组未见有骨形成。结论:将两种细胞复合支架后与背阔肌皮瓣联合应用可预构血管化骨瓣,且成骨能力与血管化程度均很理想。Objective: To vascularize bone cells and scaffold into latissimus dorsi muscle. Methods: Rabbit osteoblasts (ROB) and vascular endothelial cells (RVEC) were processed by cryopreservation, scaffold with 2.5cm × 1cm × 1cm was made and coated by collagen I , Nine Japanese rabbits were divided into three groups. Group A: ROB and RVEC in ratio of 2:1 combined with PDLLA were implanted in the rabbit left latissimus dorsi muscle, and ROB/PDLLA were implanted in the right latissimus dorsi muscle; Group B: the left was ROB + RVEC/PDLLA, the right was PDLLA; Group C: left was ROB/PDLLA, right was PDLLA. Specimens were harvested at 2w, 4w and 8w postoperatively, and examined histologyically for osteogenesis and vascularization. Results: In the group of ROB + RVEC/PDLLA, capillaries were seen running adjacent to the islands of bone tissue. The cross section of the experimental flaps after 4 and 8 weeks showed bone and vascular tissue arising in the central area. The osteogenesis ability and vascularization degree of latissimus dorsi muscle combined with co - culture of ROB and RVEC were better than that of ROB/PDLLA. Moreover there was bone formation in the group of PDLLA. Conclusion: This study demonstrates that it may be an ideal method prefabricate vascularized tissue engineered bone by co - culture of OB and VEC on latissimus dorsi muscle.
