摘　　要：目的克隆小鼠谷胺酸脱羧酶65(GAD65)基因。方法(1)用RT-PCR的方法扩增小鼠GAD65基因。(2)将GAD65克隆到T载体PUCm-T上。(3)对质粒PUCm-T/GAD65进行酶切鉴定和序列分析。结果克隆的DNA片段为1758bp的小鼠GAD65的全基因编码序列。结论成功的克隆了小鼠GAD65基因,为构建GAD65基因疫苗以及糖尿病的免疫干预治疗打下基础。Objective To construct a clone of murine GAD65. Methods （1） Mouse cDNA coding GAD65 was synthesized and amplified using RT PCR system. （2）The cDNA was then connect cd with the T vector PUCm-T. （3）The PUCm T/GAD65 clone was examined by incision enzyme and sequence analysis. Results A clone of complete murine GAD65 gene was constructed. Conclusions A clone of murine GAD65 is successfully constructed. It is a basis step for further construction of expression plasmid that expresses GAD65 and for immunologic intervention of T1DM.
