摘 要:目的:观察脑脉舒安颗粒对大鼠的长期毒性。方法:160只大鼠按体重随机分为脑脉舒安颗粒高、中、低剂量组(以生药计分别为100,50和25g·kg^(-1))及空白对照组,每组40只,雌雄各半,每日灌胃给药1次,对照组给予等体积饮用水,连续给药180 d。观察大鼠一般状况和给药90,180 d及停药后15d大鼠的血液学、血液生化学、脏器系数、组织病理学变化。结果:脑脉舒安颗粒100 g·kg^(-1)组大鼠在给药45~90 d期间,一般状况欠佳,给药90~180d时,血清胆固醇值明显低于对照组(P<0.05),停药15d后血清胆固醇值恢复正常,但组织病理学检查无异常。停药恢复15d后高、中、低剂量组大鼠体重、摄食量、生化指标检查、脏器系数、病理学检查与对照组相比均无差异。结论:脑脉舒安颗粒100g生药·kg^(-1)是大鼠的中毒剂量,25g生药·kg^(-1)为安全剂量。Objective: To evaluate long-term toxicity of the granules for cerebral tonifying and mind sedating (Naomaishuan) in rats. Methods: 160 rats were randomly supplemented with naomaishuan granules of 100, 50, 25 or g·kg^-1 daily via gastric infusion for 180 days. Each group had 40 rats, equaling gender. The general behaviors and physical status of the rats were monitored pre- and postsupplements. The hematology, blood biochemistry, organs coefficient and histopathological status of the rats were examined in 90, 180 days when supplementing and in 15 days after the termination of the supplement. Results: The rats that were dosed with 100 g·kg^-1 daily initiated poor physical conditions at 45 days post supplements and significant reductions of the serum cholesterol levels at 90 days post supplement eompared to the normal control (P 〈 0.05 ). However, the serum cholesterol levels were back to normal in 15 days after the termination of the supplements No significant histopathological abnormality was found No histopathological changes were observed in the rest of two doing groups. Conclusion: The toxic dosage of Naomaishuan granules was about 100 g·kg·d^-1 and the safe dosage about 25 g·kg^-1 in rats.
