摘 要:目的探讨枸杞多糖(LBP)对中波紫外线(UVB)辐射致体外培养人皮肤成纤维细胞损伤的保护作用。方法采用强度30mJ/cm2的中波紫外线照射体外培养的人皮肤成纤维细胞,实验分为对照组、紫外线照射模型组和四个不同浓度LBP保护组,用MTT法检测细胞增殖活性,酶生化法检测细胞内SOD活性、MDA含量,以及培养液中LDH漏出率。结果强度为30mJ/cm2的中波紫外线可以造成体外培养的人皮肤成纤维细胞增殖活性降低,细胞内SOD活性降低,MDA含量升高,LDH漏出增加,与对照组相比均P<0.05。给定浓度范围的LBP能剂量依赖性地增加细胞的增殖活性,增加细胞内SOD活性、MDA含量,减少LDH的漏出。结论紫外线可以造成体外培养的人皮肤成纤维细胞氧化损伤,LBP对UVB致人皮肤成纤维细胞的损伤具有一定保护作用,其机制可能与它的抗氧化作用和促进细胞增殖作用有关。Objective To investigate the protective effects of Lycium Barbarum Polysaccharides(LBP) on fibroblast damaged by ultraviolet B(UVB). Methods The fibroblast was irradiated by UVB (irradiation intensity:30mJ/cm^2) . The fibroblasts was randomly divided into six groups including 1 control group and 5 UVB radiated groups (radiated group, 0.1 mg/mL LBP, 0.2mg/mL LBP, 0.4 mg/mL LBP 0.8mg/mL LBP). The enzymic activities were determined by MTT methods . The content of MDA 、 the activities of intracellular SOD, and the content of LDH in the supematants of cells were determined by biochemical methods. Results Compared to controls, UVB (irradiation intensity:30mJ/cm^2) can significantly decrease the proliferation activities in fibroblast and intracellular SOD and significantly increase MDA content and LDH. In the given concentration , LBP can dose-dependently significantly improve the activities of cellular proliferation and intracellular SOD and significantly decrease the contents of intracellular MDA and the content of LDH in the supematants of cells( P 〈 0.01, respectively). Conclusion The LBP has protective effect on fibroblast irradiated by UVB. The mechanism of the protective effect mav be associated with it's effect in antioxidative and enhancing celluar proliferation.
