摘 要:目的:研究不同发育时期小鼠脑内LMO3的表达.方法:采用以SYBR Green Ⅰ为荧光染料的定量RT-PCR及以地高辛标记的cRNA为探针的原位杂交组织化学法检测LMO3基因在小鼠脑发育过程中的表达.结果:FQRT-PCR结果表明,LMO3在出生前高表达,P7d后表达水平急剧下降,至成年都维持在一个较低的水平.LMO3 mRNA在小鼠胚胎期E10.5d即有表达,但仅限于整个脑泡组织内,胚体其他区域均为阴性;E11.5d脑区信号逐渐加强,无核团及脑区的特异性,胚体其他部分仍为阴性;E15.5~P0d期LMO3阳性信号在几乎所有观察的脑区均有广泛而较强的着色;P7d LMO3阳性信号较出生前变弱,分布逐渐集中;P14~P60d LMO3mRNA表达范围较P7d逐渐缩小,但仍有广泛表达,阳性信号主要集中于不同核团,表达有强弱之分.结论:LMO3基因可能与小鼠出生前脑的发育以及出生后特定脑区神经元的特化及特性维持有关.AIM: To examine the localization of neuronal specific transcription factor LMO3 mRNA in the development of central nervous system (CNS) of the mice. METHODS : Realtime fluorescence quantitative RT-PCR( FQ RT-PCR) and in situ hybridization histochemical staining method with digoxin-labeled cRNA probe were used to detect the expression of LMO3 mRNA. RESULTS: The FQ RT-PCR showed that the expression of LMO3 mRNA was the highest during pregnancy, then it decreased significantly after birth and almost remained the same level until adult. LMO3 mRNA expression was extensive in brain at early embryonic stages (E10.5±11.5 d), but not detected at other parts of embryo. The high level in brain was still seen over mid-(E15.5d) and late- (E17.5-P0 d) embryonic brain and showed less different signal. During postnatal development, LMO3 mRNA was found gradually regionalized and showed the difference of signal in different parts of brain. LMO3 mRNA expression of P14-P60 d was still extensive although the extent was gradually more regional than that of P7d. The signal of LMO3 mRNA was mainly located in different nucleus clony and showed different intensity. CONCLUSION: LMO3 plays a role in brain development during embryonic period and postnatal differentiation and specification of specific brain regions.
