摘 要:目的:观察向小鼠小脑皮层投射的γ-氨基丁酸(GABA)能神经元在脑干内的分布.方法:四甲基罗达明(TMR)逆行追踪和免疫荧光组织化学相结合的双重标记方法,结果在激光共聚焦显微镜下观察.结果:将TMR注入小脑皮层后,TMR逆标神经元主要见于三叉神经脊束核(SP5),前庭上核(SuVe),前庭脊核(SpVe),前庭内侧核(MVe),前庭外侧核(LVe),耳蜗腹侧核(VC),耳蜗背侧核(DC),外侧上橄榄核(LSO),下橄榄主核(IOPr),下橄榄内侧核(IOM),下橄榄背侧核(IOD);GFP阳性神经元主要位于SuVe,MVe,LVe,DC,LSO,VC,SP5,SpVe,IOPr,小细胞网状核(PCRt),巨细胞网状核(Gi),在IOM和IOD内也能见到一些GFP阳性神经元;TMR/GFP双标神经元主要位于SuVe,MVe,LVe,DC,VC,SP5,SpVe,IOPr,在LSO,IOM和IOD内也能见到一些TMR/GFP双标神经元.结论:向小脑皮层投射的GABA能神经元在脑干内的分布比较广泛,它们可能参与了小脑对运动的调控.AIM: To observe the distribution of γ-aminobutyric acid (GABA)-containing neurons projecting to cerebellar cortex from the brainstem of the mouse. METHODS: Double-labeling method using tetramethyl-rhodamine (TMR) retrograde tracing combined with green fluorescent protein (GFP) immunohistochemical staining was utilized to observe the neurons under the laser scanning confocal microscope. RESULTS: TMR retrogradely labeled neurons were mainly found in nucleus of spinal tract of tfigeminal nerve (SP5), superior vestibular nucleus (SuVe), spinal vestibular nucleus (SpVe), medial vestibular nucleus ( MVe ) , lateral vestibular nucleus ( LVe ) , ventral cochlear nucleus (VC), dorsal cochlear nucleus (DC), lateral superior olivary nucleus ( LSO), principal nucleus of inferior olive ( IOPr), medial nucleus of inferior olive (IOM) and dorsal nucleus of inferior olive ( IOD ). GFP-like immunoreactive ( LI ) neurons were mainly observed in SuVe, MVe, LVe, DC, LSO, VC, SP5, SpVe, IOPr, parvocellular reticular nucleus (PCRt) and gigantocellular reticular nucleus (Gi). A few GFP-LI neurons were also found in the IOM and IOD. TMR/GFP double-labeled neurons were mainly located in SuVe, MVe, LVe, DC, VC, SP5, SpVe and IOPr. Some TMR/GFP double-labeled neurons were also observed in IOM and IOD. CONCLUSION: GABA ergic neurons projecting to the cerebellar cortex are widely located in the brainstem. These GABAergic neurons are probably involved in the coordination and integration of cerebellum for locomotion.
