摘 要:目的建立HPLC法测定人血浆中二甲双胍的浓度。方法用乙腈作为蛋白沉淀剂,色谱柱为150 mm×4.6 mm,5μm,流动相为V(乙腈):V(甲醇):V(水):V(1.5%SDS)=35:15:42.5:7.5,流速1.0 mL/min,DAD检测波长234 nm。结果二甲双胍的线性范围为0.154~2.316μg/mL,最低检测限为0.111μg/mL(s/n>3),样品平均回收率为97.8%,批内、批间精密度的RSD均小于8.0%。结论该法准确、快速、简便,可用作人血浆中二甲双胍浓度测定方法。Objective To develop a quick and accurate HPLC method for the determination of metformin in human plasma. Methods Samples of plasma were deproteinated with acetonitrile. Separation was achieved(5 μm, 4.6 mm× 150 mm). The mobile phase consisted of acetonitrile(35%), water(42.5%), methanol(15%), and sodium dodecyl sulphate(SDS, 1.5%) at a flow rate of 1.0 mL/min. Metformin was detected by DAD detector at 234 nm. Results Standard curve was linear over the range of 0.154 to 2.316 μg/mL. The detection limit in the plasma was 0.111 μg/mL(s/n 〉3), while the recovery was more than 97%. RSD of intra-batch and inter-batch were all less than 8.0%. Conclusion The method is simple, rapid, and accurate, and can be used to determine the concentration of metformin in human plasma.
