摘  要:目的:探讨BMP-7在糖尿病(DM)肾病发病机制中的作用。方法:以免疫组织化学、Westernblot及荧光实时定量PCR(RT-PCR)的方法动态观察链脲佐菌素诱导的大鼠糖尿病肾病发生过程中大鼠肾脏BMP-7蛋白及mRNA表达。结果:在正常大鼠及DM大鼠肾小管均有BMP-7蛋白表达,以胞浆表达为主,DM大鼠肾小管和集合管BMP-7蛋白表达较正常对照组明显增多(P<0·05),BMP-7mRNA的表达呈现先增加后降低的趋势。结论:作为TGF-β超家族的一员,BMP-7信号通路在糖尿病肾病发生早期可能起到重要的反馈性抑制作用。Objective: To investigate the role of bone morphogenetic protein-7 ( BMP-7 ) expression in pathogenesis of diabetic nephropathy. Methods: Fifty male Wistar rats weighing 180 to 220g were randomly divided into five groups (10 in each). Among them, four groups were diabetic nephropathy models (DM) established by a single injection of streptozotocin (STZ) at a dose of 55mg/kg body weight, while the other group ( group A ) was normal control group, of which the rats were injected with citrate buffer solution instead of STZ. In model groups, the rats were tested in 2 weeks ( group B), 4 weeks (group C ), 8 weeks (group D), and 16 weeks (group E) respectively after the injection of STZ for the expression of BMP-7 by using immunohistochemical techniques, western blot and real-time PCR. Results: BMP-7 protein expressed in nephric tubules of both DM and normal rats mainly in cytoplasma. But in nephric tubules and collectors of DM rats, the expression was significantly increased than in those of rats in control group ( P 〈 0.05 ). The expression of BMP-7 mRNA increased in 2 weeks and decreased in 16 weeks after the STZ injection. In addition, the increase of expression of of TGF-β and COL-I mRNA in diabetic rats were observed. Conclusion: As a member of TGF-β super family, BMP-7 information pathway might play an important part in feedback inhibiting diabetic nephropathy in the early stage.
